3份胡椒属种质花粉离体萌发研究

摘  要:以海南蒟、大叶蒟和球穗胡椒的花粉为材料,研究了不同培养基和采样时间对花粉萌发率的影响。结果表明:3种胡椒种质花粉的最佳萌发条件分别为100 g/L蔗糖+100 mg/L硼酸+600 mg/L Ca(NO3)2、100 g/L蔗糖+100 mg/L硼酸+300 mg/L Ca(NO3)2和100 g/L蔗糖+200 mg/L硼酸+300 mg/L Ca(NO3)2。ME3培养基抑制海南蒟和球穗胡椒花粉萌发,蔗糖、硼酸和Ca^2+的协同作用可以提高胡椒花粉萌发率,硼酸的浓度需要控制在一定范围内(≤200mg/L),300-600 mg/L的Ca^2+浓度对花粉萌发率影响不显著。同时,本研究发现,9:00-10:00或16:30是收集这3种胡椒种质花粉进行杂交试验的较佳时间。In order to investigate effect factors on pollen germination of Piper, various culture media and collecting time were performed to analyzed the germination rate with three genus Piper germplasm (Piper hainanense, Piper laetispicum, Piper thomsonii). The results indicated that the optimal culture medium for the three Piper were 100 g/L sucrose+100 mg/L H3BO3+600 mg/L Ca^2+, 100 g/L sucrose+100 mg/L H3BO3+300 mg/L Ca^2+ and 100 g/L sucrose+200 mg/L H3BO3+300 mg/L Ca^2+, respectively. The pollen germination of Piper hainanense and Piper thomsonii were suppressed by ME3 medium. The cooperation of sucrose, H3BO3 and Ca2+could promote Piper pollen germination capacity. Yet the concentration of H3BO3 should be lower than 200 mg/L. Moreover, the effect was insignificant when Ca^2+ concentration changed from 300 to 600 mg/L. For the time, it will be favorable for hybridization experiments between 9 00 and 10 00, or after 16 30.
