美洲大蠊Per a7的抗原表位特征及三维结构建模

摘  要:目的通过生物信息学方法了解美洲大蠊过敏原蛋白Per a7的结构特征,为蟑螂变态反应性疾病的诊断和治疗提供依据。方法BLAST得到Per a7相似序列,运用多种生物信息学软件构建同源进化树并预测其抗原表位及建模三维结构。结果美洲大蠊Per a7与德国小蠊精氨酸激酶在进化上具有较近的亲缘关系。Per a7主要为螺旋结构为主的非亲水性蛋白,预测其主要抗原表位集中于55—56区域。Motif预测其具有一个原肌球蛋白标志。结论运用生物信息学方法预测的三维结构基本能反应Per a7真实的空间构象,这将为今后进一步了解和掌握Per a7结构和功能的关系打下理论基础。Objective We conducted bloJnfonnatic analyzes to identify and characterize the structural properties of Per a 7, which gives the clue for the diagnosis and therapy of cockroach allergy. Methods BLAST was applied to search for the protein, which show high similarities with Per a 7. Clustal W 1.83 was used for sequence alignment and phylogenetie tree construction. Results The phylogenetic analysis found that Per a 7 showed highest similarity with the argine kinase identified from Blattella germanica.Per a 7 is mainly containing- helix secondary structure. Its antigenic epitope are mostly located on 55 - 56 regions. Motif analysis found that Per a 7 has a tropomyosins signature. Three - dimensional structure of Per a 7 was also constructed. Conclusion It seemed that the predicted structure could reflect the real tertiary structure of Per a 7, which is useful for the further study of structure - function relationship.
