摘 要:目的探讨振荡运动对负性情绪雌鼠化疗癌因性疲乏(cancer-related fatigue,CRF)的影响及相关机制。方法 60只SD雌鼠随机分为5组,对照组、CRF组、CRF+光照组、CRF+噪音组、CRF+光照+噪音组,每组12只。将2.5 mg/kg盐酸表柔比星腹腔注射到大鼠体内,累积剂量达到15 mg,并联合光照及噪音刺激建立负性情绪的癌因性疲乏动物模型,通过中等强度(25 Hz)振荡运动进行干预,每周6 d,连续8周。造模后,振荡运动末次结束后即刻用鼠尾悬挂实验评估疲乏程度,用酶联免疫吸附试验检测血清白介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)和白介素-6(IL-6)水平。结果造模后,与对照组相比,CRF组、CRF+光照组、CRF+噪音组、CRF+光照+噪音组鼠尾悬挂时间均延长(P<0.01),血清IL-1β和TNF-α水平均增高(P<0.01);与CRF组相比,CRF+光照组、CRF+噪音组、CRF+光照+噪音组鼠尾悬挂时间均延长(P<0.01),血清IL-1β和TNF-α水平均增高(P<0.01),其中以CRF+光照+噪音组最明显。振荡运动后,与运动前自身相比,CRF组、CRF+光照组、CRF+噪音组、CRF+光照+噪音组鼠尾悬挂时间均缩短(P<0.01),血清IL-1β和TNF-α水平均下降(P<0.01),IL-6水平均上升(P<0.01)。结论振荡运动能够缓解伴有负性情绪的CRF症状,IL-6可能是缓解CRF的影响因子。This study was performed to investigate the influence of vibration exercise on cancer-related fatigue(CRF) in female rats with negative emotion after chemotherapy. Total of 60 SD female rats were randomly divided into five groups, including control group, CRF group, CRF +light group, CRF +noise group and CRF +light +noise group, with 12 rats in each group. Epirubicin hydrochloride of 2.5 mg/kg was intraperitoneally injected into rat body until the cumulative dose reached 15 mg, and negative emotional stimuli of light and noise were introduced to establish negative emotion cancer-related fatigue animal model. Then moderate intensity(25 Hz) vibration exercise was given to rats six days a week for eight weeks. After modeling or the last exercise, the immobility time of tail suspension test(TST) were calculated to evaluate the status of fatigue, and enzyme linked immunosorbent assay were employed to detect serum levels of interleukin-1 beta(IL-1β), tumor necrosis factor alpha(TNF-α) and interleukin-6(IL-6). After modeling, the immobility time of TST and the serum level of IL-1β and TNF-α in CRF group, CRF+light group, CRF+noise group and CRF+light+noise group were higher than those in control group; the immobility time of TST and the serum levels of IL-1β and TNF-α in CRF+light group, CRF+noise group and CRF+light+noise group were higher than those in CRF group, and CRF+light+noise group demonstrated the most higher level. After vibration exercise, the immobility time of TST and the serum levels of IL-1β and TNF-α were lower but the serum level of IL-6 were higher in CRF group, CRF+light group, CRF+noise group and CRF+light+noise group as compared with before exercise. This study indicated that vibration exercise can alleviate the CRF accompanying with negative emotion, and IL-6 maybe plays an impact role in the processes.
