摘 要:目的研究红系衍生的核因子2相关因子2(nuclear factor-erythroid 2-related factor 2,Nrf2)在大鼠骨骼肌挫伤愈合中的表达及分布,探讨骨骼肌挫伤愈合的可能机制。方法建立大鼠骨骼肌挫伤模型,健康成年雄性Sprague-Dawley大鼠45只,体重280~320 g,随机分为9组,其中骨骼肌挫伤分为8个时间段组,另一组为正常对照组,各组5只大鼠的肌肉样本用于石蜡切片进行HE染色、免疫组织化学染色和免疫荧光染色。结果对照组大鼠骨骼肌细胞浆显示了Nrf2的弱阳性反应。骨骼肌挫伤后6~12 h,大部分多形核细胞(polymorphonuclear cells,PMNs)和少量单个核细胞(mononuclear cells,MNCs)的胞浆中呈Nrf2阳性表达。伤后1~3 d,Nrf2主要表达于MNCs的胞浆中。伤后5~14 d,Nrf2阳性反应主要见于成纤维细胞(fibroblastic cells,FBCs)的胞浆中。此外,伤后5~14 d,多核肌管和新生的毛细血管内皮细胞的胞浆也显示Nrf2阳性反应。双重免疫荧光显示在大鼠骨骼肌挫伤愈合的过程中,Nrf2时序性表达于中性粒细胞、巨噬细胞、肌成纤维细胞。结论大鼠骨骼肌挫伤愈合过程中,Nrf2表达在中性粒细胞、巨噬细胞、肌成纤维细胞、再生多核肌管的胞浆中,未在细胞核中表达,提示Nrf2可能不参与骨骼肌挫伤愈合。Objective To research the expression and distribution of nuclear factor-erythroid 2-related factor 2( Nrf2) after skeletal muscle contusion in rats,and to explore the possible mechanism of skeletal muscle wound healing. Methods A total of 45 healthy,adult SpragueDawley( SD) male rats,weighing between 280 and 320 g,were divided into control group and contusion groups. After wounding,samples were taken from the wounded sites after the animals were sacrificed after anaesthetization at 6 h,12 h,1 d,3 d,5 d,7 d,10 d and 14 d postwounding( n = 5 rats in each posttraumatic interval). The remaining 5 rats were used as controls. The specimen was used for HE staining,immunohistochemistical and immunofluorescent procedure in paraffined sections. Results In uninjured skeletal muscle specimens,a weak Nrf2 immunoreactivity was found in cytoplasm of skeletal muscles. From 6 to 12 h post-injured,Nrf2-positive staining was found in cytoplasm of most PMNs and a few MNCs. From 1 to 3 days post-injured,Nrf2 was expressed primarily in cytoplasm of MNCs,and then in FBCs during 5to 14 days. In addition,from 5 to 14 days post-injured,Nrf2-positive staining was found in cytoplasm of regenerative myotubes. Nrf2 was temporally expressed in neutrophils,macrophages and myofibroblasts throughout skeletal muscle wound healing in rats. Conclusion Nrf2 is expressed in cytoplasm of neutrophils,macrophages,myofibroblasts,endothelial cells and regeneration multinuclear myotubes during skeletal muscle wound healing,suggesting that Nrf2 may not be involved in skeletal muscle wound healing.
