摘 要:诱导获得了双季茭白龙茭2号的愈伤组织,采用RAPD和ISSR分子标记技术对4个茭白品种和诱导获得的2种茭白愈伤组织进行了基因组多态性分析。筛选获得的12条ISSR和7条RAPD引物共扩增出121条清晰条带、20条多态性条带,其中茭白品种材料与愈伤组织的多态性位点比率分别为3.30%和13.22%。正常茭白的3个品种之间未检测到多态性;正常茭白与野茭间多态性位点比率为3.31%;但愈伤组织与茭白之间的基因组多态性为11.57%。表明组培过程中产生的基因组多态性明显高于正常田间无性繁殖产生的变异,茭白组织培养体系的建立将有助于茭白的品种选育。RAPD and ISSR molecular markers were used to detect the genetic diversity of 4 varieties of Zizania latifolia and two kinds of callus induced from Longjiao 2( double cropping Zizania). 121 distinct bands and 20 polymorphic bands were amplified from the primers selected including 7 RAPD primers and 12 ISSR primers, The proportion of polymorphic loci was 3.30% and 13.22% in the Z. latifolia and callus respectively. There was no genomic polymorphism between the 3varieties of normal Z. latifolia, but 3.31% genomic polymorphism loci between normal Z. latifolia and the wild Z. latifolia.However, the genomic polymorphism loci between callus and Z. latifolia were detected to be 11.57%. Genome variation in the process of tissue culture was significantly higher than the normal field variation produced by asexual reproduction. The establishment of tissue culture system in Z. latifolia would contribute to the cultivator breeding of Z. latifolia.
