对2010年版《中国药典》中黄蜀葵花含量测定方法的改进

摘　　要：目的研究改进《中国药典》黄蜀葵花的含量测定方法,并建立同时测定黄蜀葵花中金丝桃苷、槲皮苷含量的方法。方法采用月旭XB-C18（250mm×4.6mm,5μm）色谱柱,以乙腈-0.1%磷酸溶液进行梯度洗脱,检测波长360nm,柱温30℃,流速1.0m L·min^-1。结果对含量测定进行方法学考察,结果表明均符合含量测定要求,金丝桃苷进样量在0.21~6.31μg（r=0.9998）、槲皮苷进样量在0.21~6.38μg（r=0.9995）范围内呈良好的线性关系,平均加样回收率分别为98.70%（RSD=0.92%）、97.46%（RSD=1.74%）。结论改进了《中国药典》黄蜀葵花的含量测定方法,解决金丝桃苷与相邻峰难以分离和分析时间过长的问题。并建立了同时测定金丝桃苷、槲皮苷的方法,本方法结果准确,重现性好,专属性强,可用于黄蜀葵花的质量控制。OBJECTIVE In this work,an improvement method of determination Flower of Sunset Abelmoschus in ＂Chinese Pharmacopoeia＂was developed,and a new method for determination of hyperin and quercetin in Flower of Sunset Abelmoschus was established simultaneously. METHODS A Welch UltimateXB-C18（ 25 mm × 4. 6mm,5μm） column was used for separation,with acetonitrile-0. 1% phosphoric acid as mobile phase for gradient elution. The detection wavelength was set at 360 nm,column temperature was 30 o C and flow rate was 1. 0m L · min^-1. RESULTS The method showed very good reliability for Flower of Sunset Abelmoschus determination and it can acquire the requirements of determination very well. Hyperin injection quality at 0. 21 ~ 6. 31μg（ r = 0. 9998） and quercetin injection quality at 0. 21 ~ 6. 38μg（ r = 0. 9995） showed a good linear relationship,the average recovery were 98. 70%（ RSD = 0. 92%） and 97. 46%（ RSD = 1. 74%）. CONCLUSION The experiment improved the method for determination of Abelmoschus in ＂Chinese Pharmacopoeia＂; solved the problem of hyperoside with adjacent peaks that was difficult to separate and need long analyze time. The method for simultaneous determination of hyperin and Quercitrin was established and which showed high accurate and good reproducible,and can be used for quality control in the determination of Flower of Sunset Abelmoschus.
