摘 要:【目的】探讨尼可地尔对体外培养的犬心肌细胞凋亡的影响。【方法】取成功离体培养的新生犬心肌细胞,将其随机分为对照组(A组)、过氧化氢组(B组)、尼可地尔+过氧化氢组(C组)、尼可地尔+5-羟基癸酸+过氧化氢组(D组);然后通过对一系列细胞凋亡标志物的检测,探讨尼可地尔是否具有抗细胞凋亡的作用。【结果】通过采用TUNEL染色标记、细胞色素C移位、Caspase-3活化等方法显示B组心肌细胞凋亡率明显高于C组和A组,且尼可地尔的这种抗细胞凋亡的作用可以被ATP敏感钾通道阻滞剂5-羟基癸酸所阻断。B组Caspase-3活性随时间延长而逐渐增强,与对照组相比差异具有统计学意义(P〈0.001);与B组相比,C组加入尼可地尔预处理后能够明显持续抑制Caspase-3活性,差异具有统计学意义(P〈0.001)。【结论】尼可地尔具有直接抗离体培养的新生犬心肌细胞凋亡的效果,其机制与线粒体上ATP敏感钾通道开放密切相关。[Objective]To explore the effect of nicorandil on the apoptosis of cultured neonatal dog cardiomyocytes in vitro.[Methods] The cultured neonatal dog cardiomyocytes were randomly divided into four groups: control group(group A) ; hydrogen peroxide (H2O2 ) group (group B);nicorandil plus H2O2group (group C) ; nicorandil plus 5-hydroxydecanoate (5-HD) and H2O2 group(group D). Whether nicorandil can in- hibit the apoptosis of cardiomyocytes was evaluated directly by using a number of apoptotic markers.[Results] TUNEL staining, cytochrome C translocation and Caspase-3 activation showed the apoptosis rate of group B was significantly higher than that of group C and group A, and the nicorandil anti apoptotic effect could be blocked by ATP sensitive potassium channel blocker 5- hydroxy decanoic acid. The activity of Caspase 3 in group B increased gradually with time, and the difference compared with the control group was statistically significant ( P 〈0.001) ; Compared with group B , group C was treated with nicorandil pretreatment and can significantly inhibit caspase-3 activity, and the difference was statistically significant (P 〈0.001)[Conclusions]Nicorandil has a direct anti effect on the apoptosis of cultured neonatal dog eardiomyocytes in vitro. The mechanism is closely related of mitochondrial ATP sensitive potassium openchannel.
