摘 要:细菌和古生菌利用自身的获得性免疫系统(CRISPR)抵御外来侵略,使外源DNA降解。为了基因突变兔子成纤维细胞的Myostatin基因,研究构建了CRISPR和g RNA质粒,当Cas9与g RNA摩尔数之比分别为1∶2、1∶1时进行转染到兔子成纤维细胞中,结果显示有打靶效率差异,经过鉴定均有碱基突变。结果表明:利用(CRISPR/Cas9)系统可成功突变目的基因。Bacteria and archaea used their acquiring immune system(CRISPR)to defense foreign gene and make its DNA degradation. In order to mutate myostatin gene of rabbit into fiber cells, the plasmid of CRISPR and gRNA was constructed. When the moles ratio of Cas9 and gRNA were 1:2,1:1 to transfect into rabbit fibroblasts,the results showed that there were differences in targeting efficiency after mutation identification. The results showed that the use of (CRISPR / Cas9) system could successfully mutate target gene.
