摘 要:目的果糖溶液喂养Wistar雄性大鼠,注射Wistar大鼠不同浓度的鞣云实素检测大鼠体内胰岛淀粉样多肽(IAPP)、胰高血糖素样肽-1(GLP-1)及胰岛素(INS)浓度的变化。方法 Wistar大鼠给予相同浓度果糖溶液连续灌胃3周,第4周腹腔注射不同浓度的鞣云实素溶液,隔1 d注射1次,连续注射3次,第7天取大鼠血清,利用酶标仪测定血清中胰岛淀粉样多肽、胰高血糖素样肽-1及胰岛素的浓度。结果果糖溶液喂养后胰岛素分泌减少、胰岛淀粉样多肽浓度降低、胰高血糖素样肽-1浓度升高;注射20 mg·kg-1鞣云实素溶液后胰岛素分泌增多、胰岛淀粉样多肽浓度明显降低、胰高血糖素样肽-1浓度明显增高;注射40 mg·kg-1鞣云实素溶液后胰岛素分泌与对照组相比明显减少、胰岛淀粉样多肽浓度明显增高且高于对照组、胰高血糖素样肽-1浓度明显降低。结论低浓度鞣云实素能够促进胰高血糖素样肽-1生成、刺激胰岛素分泌及抑制胰岛淀粉样多肽的形成,本实验为进一步研究鞣云实素对胰岛素抵抗大鼠模型的相关激素变化提供一定基础。Objective To feed male Wistar rats with fructose solution,and inject different concentration of corilagin so-lution to Wistar rats,determine the changes of the concentration of IAPP,GLP - 1)and INS in rats serum. Methods To give same concentration of fructose solution to Wistar rats three weeks by intragastric administration. In the fourth week,dif-ferent concentration of corilagin solution was admined by intraperitoneal injection,once every two days and three times. In the seventh day,blood serum of rats was collected and the concentrations of IAPP,GLP - 1 and INS were determined by ELISA. Results After fed fructose solution,the rats secreted insulin was decreased,the concentration of IAPP was reduced and the concentration of GLP - 1 was increased;After injected the concentration of 20 mg·kg - 1 corilagin,the insulin secre-tion was increased,the concentration of IAPP was decreased significantly,the concentration of GLP - 1 was increased signifi-cantly;When injected the concentration of 40 mg·kg - 1 corilagin,the insulin secretion was decreased significantly to com-pared with control group,the concentration of IAPP was increased significantly and higher than control group,the concentra-tion of GLP - 1 was reduced significantly and lower than control group. Conclusion Corilagin at low concentration can pro-mote the generation of GLP - 1,stimulate insulin secretion and inhibit the formation of IAPP. The experiment provided some basis on the further research of corilagin for the hormonal changes related to insulin resistance in rats model.
