四神丸对溃疡性结肠炎模型大鼠Toll样受体2、白细胞介素-1β和白细胞介素-4的影响

摘　　要：目的观察四神丸对溃疡性结肠炎(UC)模型大鼠结肠组织Toll样受体2(TLR2)基因表达及血清白细胞介素(IL)-1β、IL-4含量的影响,探讨其治疗UC的作用机制。方法采用复合方法制作脾肾阳虚型UC大鼠模型。实验大鼠分为空白组、模型组、阳性药组、四神丸组,阳性药组予柳氮磺吡啶肠溶片水溶液0.5 g/kg灌胃,四神丸组予四神丸26 g/kg灌胃,每日1次,共21 d。肉眼观察结肠组织损伤情况,并检测大鼠结肠组织TLR2基因表达及血清IL-1β、IL-4的含量。结果与空白组比较,模型组大鼠结肠黏膜肉眼观察有显著炎症、充血、溃疡形成(P<0.01),模型组大鼠结肠组织TLR2基因表达与血清IL-1β含量升高(P<0.05),IL-4含量降低(P<0.01);与模型组比较,四神丸组大鼠结肠组织TLR2基因表达和血清IL-1β含量均降低(P<0.05,P<0.01),IL-4含量升高(P<0.05)。结论四神丸治疗UC机制可能与调节肠道免疫细胞因子、抑制TLR2信号避免其过度激活有关。Objective To observe the effects of Sishen Pill on expressions of TLR2, IL-1β and IL-4 in ulcerative colitis model rats;To investigate the mechanism of Sishen Pill for the treatment of ulcerative colitis. Methods Compound method was used to establish spleen-kidney-yang-deficiency ulcerative colitis model rats. Experimental rats were divided into blank group, model group, positive medicine group, and Sishen Pill group. Positive medicine group was given SASP aqueous solution 0.5 g/kg for gavage, while Sishen Pill group was given Sishen Pill 26 g/kg for gavage. The damage in colon tissue was observed with naked eyes. The expressions of TLR2 in colon tissue and IL-1β and IL-4 in serum were detected. Results Compared with the blank group, rats in the model group showed formation of inflammation, congestion and ulcer under general state and colonic mucosa visual inspection(P <0.01);TLR2 in colon tissue and IL-1β increased in serum(P <0.05);IL-4 level decreased(P <0.01). Compared with model group, TLR2 in colon tissue and IL-1β inseerum decreased significantly in the Sishen Pill group(P <0.05, P <0.01);IL-4 level increased(P <0.05). Conclusion The mechanism of Sishen Pill may be related to regulating colon immune cytokines and controlling TLR2 signal to avoid activating excessively.
