摘 要:目的探讨替米沙坦对噪声暴露大鼠心肌NADPH氧化酶表达的影响,为研究噪声损伤心肌的机制提供依据。方法雄性SD大鼠随机分为正常组(C)、噪声组(N)、替米沙坦干预组(T),每组6只。N组和T组暴露于100 d B(SPL)白噪声,6 h/d,连续12周。T组每日盐酸替米沙坦水溶液灌胃〔10 mg/(kg·d)〕。采用比色法检测心肌丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性和总抗氧化力(T-AOC),采用实时荧光定量RT-PCR法测定心肌组织NADPH氧化酶亚基p47phox、p22phox、gp91phox mRNA表达,采用western blot法检测p47phox蛋白表达。结果与C组相比,N组心肌MDA含量、p47phox mRNA、p22phox mRNA、gp91phox mRNA表达水平均明显升高(P<0.05,P<0.01),T-AOC明显降低(P<0.01);与N组相比,T组心肌MDA含量、p22phox、gp91phox mRNA表达水平明显降低(P<0.05,P<0.01),SOD活性明显升高(P<0.01)。结论替米沙坦可降低NADPH氧化酶的表达,减小氧化应激,从而减轻噪声对心肌的损害。Objective To study the effects of telmisartan on the expression of NADPH oxidase of myocardium in rats exposed by noise,so as to provide the basis for the mechanism of myocardium damage induced by noise exposure.Methods Male SD rats were divided into 3 groups:the control group( C group),the noise exposure group( N group),the noise exposure plus telmisartan group( T group),with 6 rats per group.The N group and the T group were exposed to the white noise( 100 d B( SPL),6 h / day,12 weeks).The T group were administered telmisartan( 10 mg / kg per day).Malondialdehyde( MDA) level,superoxide dismutase activity( SOD),total antioxidant capacity in ventricular muscle were measured by colorimetry.p47phox、p22phox、gp91phox mRNA of NADPH oxidase subunits were determined by Real-time RT-PCR.Cardiac protein expression of p47 phox was determined by western blot.Results MDA level,expression of p47 phox mRNA,P22 phox mRNA and gp91 phox mRNA in N group were significant higher than those in C group( P〈 0.05 or P〈 0.01).T-AOC in N group was lower than that in C group( P〈 0.01),protein expression of p47 phox in N group was 0.67 ± 0.19,which was significant higher than that in C group( 0.22± 0.05,P〈 0.01).MDA level and expression of p22 phox mRNA,gp91 phox mRNA in T group were significant lower than those in N group( P〈 0.05 or P〈 0.01).SOD activity in T group was higher than that in N group( P〈 0.01).Conclusion Telmisartan treatment can decrease the expression level of NADPH oxidase in myocardium,reduce oxidative stress sources,and thus alleviate myocardium damage induced by noise exposure.
