摘  要:目的探讨太原市冬季大气PM2.5暴露对小鼠体内各脏器重量及炎症因子表达的影响。方法于2013年1月14日至2月14日在山西大学某楼顶采集大气PM2.5样品。将20只SPF级6周龄雄性小鼠随机分为暴露组和对照组各10只,其中染毒组每次以鼻滴入法给予20μl 6 mg/ml的PM2.5溶液,每2 d给药1次,连续30 d;对照组以鼻滴入法给予等量的生理盐水。解剖小鼠并称量支气管、心脏、脾、肺、肾、脑、胃重量,采用实时荧光定量PCR法检测各脏器炎症因子的相对表达。结果染毒组小鼠的支气管、脾、肺、肾、脑、胃组织脏器系数低于对照组,差异有统计学意义(P<0.05)。染毒组小鼠脾和脑中炎症因子IL-1β、IL-6、COX-2、TNF-α、NF-κB的表达高于对照组,肾IL-1β、COX-2、NF-κB的表达高于对照组,肺IL-1β、IL-6、COX-2、NF-κB的表达均高于对照组,差异均有统计学意义(P<0.05)。结论大气PM2.5暴露可减轻小鼠脏器重量,上调炎症因子的表达。Objective To investigate the effects of PM2.5 collected in winter of Taiyuan on the weights of various organs and expressions of inflammatory cytokines in mice. Methods PM2.5 samples were collected on the rooftop of a building in Shanxi University from January 14 to February 14, 2013. SPF-grade male SD mice were randomly divided into two groups( n =10),including control and PM2.5 group. The animals received either 20 μl physiological saline or PM2.5 diluted in sterile PBS(6 mg/ml) by intranasal instillation, which were performed one time every two days for consecutive 30 days. Then the weights of bronchus, heart, spleen, lung, kidney, brain and stomach were measured. RT-PCR assay was carried out to determine the expressions of inflammatory cytokines in the organs. Results In exposure group, the organ coefficients of lung, spleen, lung,kidney, brain and stomach were lower compared with the control group(P〈0.05), and the expression of inflammatory factor IL-1,IL-6, COX-2, TNF-α, NF-κB in spleen and brain of the exposure group was higher than that in control group, the expression of IL-1, COX-2 and NF-κB was higher than that in control group, the expression of IL-1, IL-6, COX-2, NF-κB in lung was higher than that in control group(P 〈0.05). Conclusion PM2.5 collected from Taiyuan in winter may decrease the organ weights and increase inflammatory cytokine expressions in mice.
