摘 要:目的探讨强直性脊柱炎(AS)患者与健康患者外周血单核细胞(PBMC)表面的TLR2和TLR4表达差异及临床意义。方法根据入选标,LM准选取40例强直性脊柱炎患者和40例健康体检患者。在未给予系统治疗前提取外周血单核细胞进行研究。并对患者红细胞沉降率(ESR)和血浆C-反应蛋白(CRP)进行检测。使用qPCR法对外周血单核细胞TLR2和TLR4mRNA表达水平进行检测。并使用western blot法对外周血单核细胞TLR2和TLR4蛋白表达水平进行检测。结果结果显示强直性脊柱炎患者ESR和CRP较对照组明显升高,差异均有显著统计学意义(P均<0.05)。同时我们还发现强直性脊柱炎患者外周血单核细胞(PBMC)TLR2和TLR4mRNA和蛋白表达水平较健康体检患者显著增加,差异均有显著统计学意义(P均<0.05)。结论 TLR2和TLR4在强直性脊柱炎患者外周血单核细胞表面的表达水平呈显著性升高,提示TLR2和TLR4与强直性脊柱炎发病可能存在密切的相关性。TLR2和TLR4表达水平有作为诊断强直性脊柱炎的潜在价值,但仍需进一步的研究证实。Objective To evaluate the value of TLRZ and TLR4 expression in peripheral blood monouclear cell (PBMC) in patients with ankylosing spondylitis (AS) and its potential meaning. Methods 40 patients with ankylosing spondylitis (AS) and forty healthy controls were enrolled in our current study. Before any systemic treatments, peripher- al blood monouclear cells (PBMC) in both groups were obtained. Meanwhile, erythrocyte sedimentation rate (ERS) and the serum level of C-reaction protein (CRP) were measured. Then, the mRNA and protein expression of TLRZ and TLR4 in PBMC were analyzed by qPCR and western blot, respectively. Results Our data figured out that ESR and CRP in patients with AS were much higher than in healthy controls. Furthermore, we also found that the mRNA and protein expression of TLR2 and TLR4 in PBMC in patients with AS were markedly up-regulated, compared with healthy controls. Conclusion These data suggests that TLR2 and TLR4 played the critical roles in immune response of ankylo- sing spondylitis. TLR2 and TLR4 expression in peripheral blood monocytes probably can be used as a marker of ankylo- sing spondylitis. However, more studies should be carried out to further evaluation.
