摘 要:目的:优选飞龙掌血总生物碱的提取工艺。方法:以酸性染料比色法测定的总生物碱含量为指标,采用单因素和正交试验法,考察液料比、乙醇体积分数、提取温度对提取工艺的影响,确定最佳提取工艺。结果:各因素对乙醇回流提取工艺的影响顺序为提取温度、液料比、乙醇体积分数,最佳提取工艺为用25倍量75%乙醇,75℃回流提取40 min。结论:优选的提取工艺稳定可行,提取率高,为飞龙掌血资源的充分利用提供实验依据。Objective:To optimize the extraction process of total alkaloids in Toddalia asiatica. Methods:With extraction amount of total alkaloids determined by acid dye colorimetry as index,effect of solid-liquid ratio,ethanol concentration,extraction temperature,the best process was screened by single factor and orthogonal experimental design. Results:The influencing order of each factor on ethanol reflux extraction process was extraction temperature,solid-liquid ratio,ethanol concentration. The optimum extraction process is as following:extracted 40 min with 25 times the amount of 75% ethanol at 75 ℃. Conclusion:Optimized technology is stable and feasible with high extraction rate,and it provides the experimental basis for full use of Toddalia asiatica.
