保肝消脂汤对非酒精性脂肪肝大鼠脂代谢及肝功能的影响

摘　　要：目的:探讨保肝消脂汤对非酒精性脂肪肝(NAFLD)大鼠脂代谢及肝功能的影响。方法:60只大鼠随机分为6组,正常组,模型组,保肝消脂汤低、中、高剂量组(1.65、3.3、6.6 g/kg),东宝肝泰(0.09 g/kg蛋氨酸)组,除正常组给予普通饲料外,其余5组都以高脂饲料喂养,造模同时连续灌胃给予各组药物,正常组及模型组给予等量生理盐水。8周后,处死大鼠,主动脉取血,检测各组大鼠血清中总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)及高密度脂蛋白胆固醇(HDL-C)含量;制备肝匀浆,检测各组大鼠肝匀浆组织中TC、TG、谷丙转氨酶(ALT)及谷草转氨酶(AST)含量;HE染色检测肝脏病理学形态。结果:与模型组比较,保肝消脂汤低、中、高剂量组均能降低模型大鼠血清中TC、TG、LDL-C及FFA的含量,升高HDL-C含量,能降低肝匀浆组织中TC、TG、ALT及AST含量,并能减轻肝脏脂肪变性及炎症。结论:保肝消脂汤能降低高脂血模型大鼠血脂水平,能保护肝功能,并减轻肝脏脂肪变性及炎症,起到治疗大鼠脂肪肝的作用。Objective： To observe the effect of Cellulite Reducing Decoction on liver lipid metabolism and liver function in nonalcoholic fatty liver（ NAFLD） rats. Methods： The rats were randomly divided into normal control group,model group,Cellulite Reducing Decoction high,medium and low dose groups（ 1. 65,3. 3,6. 6 g / kg）,Dongbao Gantai group（ 0. 09 g / kg methionine）. The normal group was given the ordinary feed and the other five groups were fed with high fat diet. Each group was given the corresponding medicine by intragastric way and the control and model groups were given normal saline for 28 d successively. Rats were executed and the blood was isolated from abdominal aorta. The concentrations of total cholesterol（ TC）,triglyceride（ TG）,low density lipoprotein cholesterol（ LDL- C）,high density lipoprotein cholesterol（ HDL- C） in serum were measured. Preparation of liver homogenate was to detect the contents of TC,TG,alanine aminotransferase（ ALT）,and aspartate aminotransferase（ AST）in liver homogenate. HE staining method was used to examine the pathology morphology of liver. Results： Compared with model group,the levels of TC,TG,LDL- C in serum and TC,TG,ALT,AST in liver tissue were decreased in Cellulite Reducing Decoction high,medium and low dose groups. The level of HDL- C was increased in Cellulite Reducing Decoction high,medium and low dose groups. Moreover,Cellulite Reducing Decoction relieved liver fatty degeneration. Conclusion： Cellulite Reducing Decoction could reduce high fat blood,protect the liver function and inhibit hepatic steatosis and inflammation,playing a role in treatment of fatty liver in rats.
