不同提取方法大黄水提取物对幼年大鼠子宫Bcl-2和Bax表达的影响

摘　　要：目的:观察不同提取方法(煎煮和水浸)大黄水提取物对幼年雌性大鼠子宫Bcl-2和Bax蛋白表达的影响。方法:4周龄雌性SD大鼠随机分为正常对照组,大黄水浸剂0.5、1.0、2.0g/kg组和大黄水煎液0.5、1.0、2.0g/kg组,每组10只,各用药组大鼠连续灌胃给药30d。采用SP免疫组化法检测大鼠子宫Bcl-2和Bax蛋白的表达情况。结果:不同提取方法大黄水提物对雌性幼年大鼠子宫Bcl-2和Bax蛋白表达均有不同程度的影响。与正常对照组比较,水浸剂组和水煎液组大鼠子宫Bcl-2蛋白表达显著降低、Bax蛋白表达显著升高;并且,水煎液各剂量组大鼠子宫Bcl-2蛋白表达明显低于、Bax蛋白表达明显高于水浸剂相同剂量组,其中水煎液2.0g/kg剂量组大鼠子宫Bcl-2和Bax蛋白表达的变化最为明显。结论:大黄水提取物对幼年雌性大鼠子宫Bcl-2和Bax有一定影响,其影响程度与提取方法和剂量有关。Objective： To observe the effects of water extract of Rhubarb by different extraction methods on expression of Bcl-2 and Bax protein in young female rats uterus. Methods： 4-week-old female SD rats were randomly divided intonormal control group, Rhubarb water agent 0.5, 1.0, 2.0g/kg groups and Rhubarb water decoction 0.5, 1.0, 2.0g/kg groups with 10 rats in each group. The rats in each drug group were lavaged with corresponding water extract of Rhubarb for 30 days. SP immunohistochemical stain was used to detect the expression of Bcl-2 and Bax protein in uterus. Results： Water extract of Rhubarb by different extraction had different effects on Bcl-2 and Bax protein expression in young rats uterus. Compared with rats in normal control group, the Bcl-2 protein expression in uterus of rats in Rhubarb water agent groups and Rhubarb water decoction groups decreased obviously, while the Bax protein expression increased obviously. Moreover, the Bcl-2 protein expression in uterus of rats in each Rhubarb water decoction group was obviously lower than that of rats in each Rhubarb water agent group, while Bax protein expression was obviously higher. Among 3 Rhubarb water decoction groups, the changes of Bcl-2 and Bax protein expression in uterus of rats in 2.0g/kg group was the most. Conclusions： Water extract of Rhubarb has certain effects on Bcl-2 and Bax expression in young female rats uterus, which is concerned with extraction methods and dose.
