摘 要:目的研究Neuroligin1基因在Wistar大鼠胚胎的肠神经系统(ENS)中的表达情况,探讨Neuroligin1基因的表达与ENS和ENS疾病之间的关系。方法取胚胎第16(E16)、18(E18)、20(E20)天和出生第一天(B1)的胚鼠肠管标本各30只,通过双重免疫荧光染色、Western blot、实时荧光定量PCR(Quantitative Real—timePCR,qRT-PCR)等实验方法,研究Neuroligin1基因在Wistar大鼠胚胎ENS各个发育时期中的表达情况。结果在Wistar大鼠E16、E18、E20和B1四个ENS发育时期中:双重免疫荧光染色显示Neuroligin1基因阳性面积的平均光密度值分别为E16(0.046±0.031),E18(0.146±0.033),E20(0.233±0.034),B1(0.345±0.344),且四组之间的差别有统计学意义(F=441.614,P〈0.05);Western blot显示Neuroligin1基因的平均蛋白表达量分别为E16(0.053±0.025)、E18(0.146±0.024)、E20(0.249±0.034)、B1(0.533±0.049),且四组之间的差异有统计学意义(F=1070.296,P〈0.05);qRT-PCR显示Neuroligin1基因的mRNA相对平均浓度分别为E16(2.543±1.294)、E18(4.637±1.830)、E20(6.814±1.552)、B1(8.345±1.364),且四组之间的差异有统计学意义(F=81.440,P〈0.05),同时qRT-PCR试验中Neuroligin1基因的mRNA表达趋势与Western blot中Neuroligin1基因的蛋白表达趋势一致。结论在E16到B1这段时期内,随着Wistar大鼠ENS的发育,Neuroligin1在ENS的表达呈递增趋势;Neuroligin1在ENS的异常表达发生越早,相关疾病可能就越严重;Neuroligin1基因相关的ENS疾病可能是先天发育异常。Objective To explore the expression pattern of neuroliginl in enteric nervous system (ENS) of rats and elucidate the relationship between neuroliginl and ENS or related diseases. Methods Intestinal tissues were harvested from 30 Wistar embryonic rats at the timepoints of 16th day pregnancy (El6), 18th day pregnancy (El8), 20th day pregnancy (E20) and 1st birth (B1). Doubleimmunofluorescent labeling, Western blot and quantitative real-time PCR (qRT-PCR) were used for observing the expression of neuroliginl during each period. Results During the periods of El6, El8, E20 and B1, double-immunofluorescent labeling showed that the average optical density of positive area was El6(0. 046± 0. 031), E18(0. 146 ± 0. 033), E20(0. 233 ± 0. 034), BI(0. 345 ±0. 344). And their differences had statistical significance (F = 441. 614, P〈0. 05). Western blot showed that the average protein expression was El6(0. 053 ± 0. 025), El8(0. 146± 0. 024), E20(0. 249 ± 0. 034) and B1 (0. 533 ± 0. 049). And their differences had statistical significance (F= 1070. 296, P〈0. 05). qRT- PCR indicated that the comparative mean concentrations of mRNA was E16 (2. 543 ± 1. 294), El8 (4. 637± 1. 830), E20(6. 814 ± 1. 552) and B1 (8. 345 ± 1. 364). And their differences had statistical significance (F = 81. 440, P〈0. 05). At the same time, the expression trend of mRNA during four periods agreed with the results of Western blot. Conclusions During the periods of E6 to B1, neurotiginl increases gradually along with ENS growth in rats. The earlier onset of an abnormal expression of neuroliginl, the greater severity of correlative diseases. And ENS diseases related with neuroliginl are mostly congenital developmental malformations.
