摘　　要：目的应用PLG-CD4方法检测HIV/AIDS全血的CD4+T绝对计数,与传统流式细胞仪比较其准确性;分析在不同时间检测结果的重复性。方法采集102份HIV感染者的抗凝全血,在采血的d 1用BD Calibur流式细胞仪的CD4/CD8/CD3和PLG-CD4方法检测CD4+T绝对计数;其中43份样本采用PLG-CD4方法在1、3、5、7 d检测结果的重复性分析;将43份样本应用PLG-CD4方法检测的d 1的结果分别与d 3、d 5、d 7的结果进行准确性的比较。结果对102份标本用BD Calibur和PLG-CD4方法检测CD4+T绝对计数进行相关性比较,R=0.908,R大于0.9,有高度相关性;对43份样本在1、3、5、7 d检测的CD4+T绝对计数进行重复性的比较分析,P=0.197,大于0.05,说明实验的组间数据无统计学差异;PLG-CD4在d 1检测结果分别与3、5、7 d的做两两之间比较,F值为0.855(F>0.05)、0.112(F>0.05)、0.002(F<0.05)。结论 PLG-CD4方法与BDCalibur的CD4/CD8/CD3方法比较,样本保存5 d内的检测结果有高度的相关性和准确性。Objective The aim of this study was to test the absolute CD4 T lymphocyte count from HIV/AIDS patients＇ whole blood specimens by PLG-CD4 technology, compare the accuracy with those detected by the traditional flow cytometry and analyze the reproducibility during different time intervals. Methods One hundred and two specimens were detected with traditional CD4/CDS/CD3 technology and PLG-CD4 technology using flow cytometry in the first day （d1） of the blood collection. And 43 out of 102 specimens were also detected by PLG-CD4 technology in d3, d5 and dT, respectively. The reproducibility and accuracy were analyzed. Results The results of CD4 T cell counts tested by two technologies showed high correlation （R=0.908）. By analyzing the variance of the CD4 T cell counts during the different time intervals （dl, d3, d5 and d7）, and the results indicated that there was no significant difference between four time periods （P=0.197）. The CD4 T cell counts in dl tested by PLG-CD4 were compared with those tested in other three time intervals （d2, d5 and d7） respectively, and F values were 0.855 （F〉0.05）, 0.112 （F〉0.05）, and 0.002 （F〈0.05）. Conclusion The CD4 T cell counts detected by PLG-CD4 technology showed high correlation and accuracy compared with the CD4/CDS/CD3 technology for the bold specimens collected among five days.
