摘  要:目的观察葛花解酲方对肝癌细胞原位移植瘤小鼠抗氧化作用的影响。方法采用H22小鼠腹水瘤细胞原位移植瘤动物模型,对癌变小鼠进行治疗。通过HE染色切片观察小鼠肝脏的组织和细胞形态变化。ELISA检测血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)的含量并观察免疫组化染色的表达情况。结果 H22腹水瘤细胞诱发的原位移植瘤小鼠肝脏可出现无限增殖、坏死等病理变化。葛花解酲方可调节SOD、MDA、GSH-Px的表达水平,在一定程度上具有抗氧化损伤作用,进而减缓模型小鼠癌变进程。结论葛花解酲方对H22小鼠腹水瘤细胞诱发的肝脏肿瘤有确切的疗效,其机制可能与抗氧化作用的激活有关。Objective To observe the antioxidant effects of Gehua JieCheng Fang ( Kudzuvine Flower Formula for hang-over symptoms) on the orthotopic allograft mice.Methods Orthotopic allograft models were established using H22 ascites tumor cells.Cancerous mice were then treated.Pathological changes of the hepatic lobe were microscopically observed after HE staining.Levels of superoxide dismutase ( SOD ) , Glutathione peroxidase ( GSH-Px ) , and Malondialdehyde ( MDA ) in the serum were determined by using ELISA methods.Expression levels of SOD, GSH-Px, and MDA in hepatic tissues were compared.Results Liver of the orthotopic allograft mice induced by H22 ascites tumor cells presented with pathological changes of unlimited proliferation and necrosis. Gehua JieCheng Fang affected the expression of SOD, MDA, and GSH-Px, which demonstrated the antioxidant effects.It could slow down carcinogenesis in these mice.Conclusion Gehua JieCheng Fang seems to have therapeutic effects on mice with hepatocarcinoma induced by H22 ascites cells.Its mechanism is related to the activation of antioxidation.
