多西紫杉醇对肝细胞裸鼠皮下移植瘤细胞生长与凋亡的作用

摘　　要：目的 探讨多西紫杉醇对肝癌细胞株裸鼠皮下移植瘤生长的影响及其可能的机制.方法 建立肝癌细胞株SMMC-7721裸鼠皮下移植瘤模型,建模成功后,将20只裸鼠分成2组,多西紫杉醇组及肿瘤对照组各10只.实验组用多西紫杉醇处理,50 μg＆#183;0.3 mL-1＆#183;只-1,每3天注射1次,连续注射8次.对照组注射生理盐水,剂量同前.在此期间,观察2组的肿瘤生长情况.30 d后切取瘤样组织,称重及测量体积,并计算抑瘤率.用TUNEL法检测肿瘤细胞凋亡,免疫组化法检测hTERT、Ki 67阳性表达率.结果 实验组的肿瘤生长速率明显减慢,实验组的肿瘤体积和瘤重明显低于对照组,两者间差异有统计学意义（P＜0.05）,其抑瘤率有效;实验组的凋亡指数明显高于对照组,两者间差异有统计学意义（P＜0.05）.实验组hTERT、Ki-67阳性表达率明显低于对照组.结论 多西紫杉醇对肝细胞癌裸鼠移植瘤具有生长抑制作用,可能与诱导凋亡及抑制增殖有关.Objective To investigate the effect of docetaxel on the growth of transplantation tumor of human hepatoma cell line in nude mice subcutaneous and its possible mechanism. Methods 20 nude mice were divided into two groups,randomly,the docetaxel group and the tumor group, 10 mice in each group after the success of modeling of hepatocellular carcinoma cell line SMMC-7721 in nude mice tumor model. The experiment group was treated with docetaxel andeache mice was injected 50μg/0.3 mL,every 3 days for 8 consecutive times. The control group was injected with physiological saline with the same dose. During this period, tumor growth was observed in two groups. 30 days later, the mice＇s tumor were excised to calculate tumor inhibition rate of each group by weighing and measuring the volume. TUNEL was used to detect the apoptosis of tumor ceils and immunohistochemical method was used to detect the expression rate of hTERT and Ki-67. Results The docetaxel of tumor growth rate obviously slowed down and the volume of the tumor was significantly lower than the control group. The difference was statistically significant （all P〈0.05 ）, which means the inhibition rate was effective. Apoptosis index in the edocetaxel group was significantly higher than that in the control group,and the positive expression rate of hTERT, Ki67 in experimental group was significantly lower than the control group （all P〈0.05）. Conclusion Docetaxel can inhibit the growth by inducing apoptosis of hepatocellular carcinoma and inhibiting cell proliferation in nude mice possiblely.
