冠心止痛胶囊含药血清对过氧化氢损伤的人脐静脉内皮细胞的保护作用

摘　　要：目的观察冠心止痛胶囊含药血清对过氧化氢(H2O2)诱导人脐静脉内皮细胞(HUVEC)损伤的保护作用。方法体外培养HUVEC,分为空白对照组、模型对照组、冠心止痛胶囊含药血清低、中、高浓度组(体积分数为5.00%、10.00%、15.00%),显微镜观察细胞形态;MTT法检测细胞活力,微板法检测上清液中乳酸脱氢酶(LDH)的活性及一氧化氮(NO)的含量,ELISA法检测内皮素-1(ET-1)、组织型纤溶酶原激活物(t PA)、纤溶酶原激活物抑制剂(PAI-1)的含量。结果与模型对照组比较,冠心止痛胶囊中、高浓度组细胞活力显著升高,LDH活性显著减低、ET-1和PAI-1含量显著减少,NO和t PA显著升高(P<0.05或P<0.01)。结论冠心止痛胶囊含药血清对H2O2诱导的HUVEC损伤具有显著地保护作用。Objective： To observe the protective effect of medicated serum prepared with Guanxin Zhitong Capsule on the injury of human umbilical veinendothelial cell（HUVEC） injured by hydrogen peroxide. Methods：The HUVEC were cultured vitro and divided into five groups as follows ： the control group,the model group,the low-dosage of Guanxin Zhitong Capsule group,the mid-dosage of Guanxin Zhitong Capsule group and the highdosage of Guanxin Zhitong Capsule group（volume fraction was 5%,10%,15%）. Cell morphology was observed under inverted phase contrast microscope. MTT was used to determine the HUVEC viability. The contents of lactate dehydrogenase（LDH）,nitric oxide（NO） were measured with the microplate and the method of enzyme-linked immunosorbent assay（ELISA） was used to determine endothelin-1（ET-1）,tissue plasminogen activator（t PA） and plasminogen activator inhibitor-1（PAI-1）. Results： Compared with the model group,the mid-dosage of Guanxin Zhitong Capsule and high-dosage of Guanxin Zhitong Capsule significantly increased HUVECs viability,decreased the contents of LDH,ET-1,PAI-1 and increased the contents of NO and t PA（P〈0.05 or P〈0.01）. Conclusion： Medicated serum prepared with Guanxin Zhitong Capsule has significant protective effect on HUVEC injured by H2O2.
