平哮合剂对过敏性哮喘豚鼠模型血清和肺泡灌洗液IL-5及IL-10水平的影响

摘　　要：目的研究平哮合剂对过敏性哮喘豚鼠模型血清和肺泡灌洗液(BALF)中白细胞介素5(IL-5)及白细胞介素10(IL-10)水平的影响。方法取豚鼠50只,以腹腔注射10%卵蛋白溶液制作过敏性哮喘豚鼠模型,后取40只造模成功豚鼠,随机分为4组,即模型对照组、平哮合剂1组、平哮合剂2组及地塞米松组,每组各10只,另取未进行造模的豚鼠10只为空白对照组。模型对照组及空白对照组均予0.9%氯化钠注射液5 m L/kg灌胃,平哮合剂1组、平哮合剂2组分别予平哮合剂5.0、2.5 m L/kg灌胃给药,地塞米松组予醋酸地塞米松0.1 mg/kg灌胃给药,均每日1次,连续7 d。检测5组豚鼠血清和BALF中IL-5及IL-10含量。结果与空白对照组比较,模型对照组豚鼠血清和BALF中IL-5水平显著升高(P<0.01),IL-10水平显著降低(P<0.01)。与模型对照组比较,平哮合剂1组、平哮合剂2组及地塞米松组血清和BALF中IL-5水平显著降低(P<0.01,P<0.05),IL-10水平显著升高(P<0.01,P<0.05)。平哮合剂1组、平哮合剂2组及地塞米松组血清及BALF中IL-5和IL-10水平两两比较差异均无统计学意义(P>0.05)。结论平哮合剂能通过作用于细胞因子IL-5及IL-10达到治疗哮喘的作用,并可能呈现一定的量效关系,为中医药治疗哮喘提供了现代实验依据。Objective To study the influence of Pingxiao mixtures on the interleukin -5 （IL -5） and interleukin- 10 （IL- 10） of serum and bronchoalveolar lavage fluid （BALF） in allergic asthma guinea pigs model. Methods Totally 40 allergic asthma guinea pigs models of 50 guines pigs were successfully induced by peritoneal injection of 10% ovalbumin （OVA） solution, and 40 allergic asthma guinea pigs were randomly divided into model control group, Pingxiao mixtures one group, Pingxiao mixtures two group and dexamethasone group, 10 guinea pigs in each group. Chosed another 10 guinea pigs which were not molding into blank control group. Guinea pigs in model control group and blank control group were received 0. 9% sodium chloride injection （5 mL/kg） by gavage. Guinea pigs in Pingxiao mixtures one and two group were received Pingxiao mixtures 5.0 ml/kg and 2.5 ml/kg by garage re- spectively. Guinea pigs in dexamethasone group were received dexamethasone acetate （0.1 mg/kg） by garage. All guinea pigs received injection once daily for 7 d. The contents of IL - 5 and IL - 10 in serum and BALF were detected in five groups. Results As compared with the blank control group, the level in IL -5 in serum and BALF were obviously increased in model control group （ P 〈 0.01 ） , and the level of IL - 10 was obviously decreased （ P 〈 O, 01 ）. As compared with model control group, the level of IL -5 in serum and BALF were obviously decreased in Pingxiao mixtures one group, Pingxiao mixtures two group and dexamethasone group （P 〈 0.05, P 〈 0.01 ）, and the level of IL - 10 was obviously increased （ P 〈 0. 05, P 〈 0.01 ）. In Pingxiao mixtures one group, Pingxiao mixtures two group and dexamethasone group, there were no statistical significant differences on the levels of IL -5 and IL - 10 in serum and BALF （ P 〉 O. 05 ）. Conclusion Pingxiao mixtures can treat asthma by acting on the IL - 5 and IL - 10, provide the modern experimental basis for the traditional Chinese medicine therapy on the
