摘 要:【目的】通过ISSR分析标记技术,分析新疆野杏种质资源的遗传多样性,为该资源的利用和保护提供理论依据和技术支持。【方法】利用ISSR分子标记对新疆伊犁霍城县大西沟的61份野杏单株进行遗传多样性分析。【结果】用21条引物对样品进行PCR扩增,共扩增出437条条带,其中多态性条带408条,多态性条带比率为92.46%。通过软件计算出有效等位基因数(Ne)、Nei’s基因多样性指数(H)和Shannon信息多样性指数(I)分别为1.5775、0.3341、0.4990。引物平均多样性信息指数(PIC)为0.91。说明野杏的遗传多样性较为丰富。利用NTSYS—PC2.1软件进行UPGMA聚类,各个样品间的遗传相似系数在0.62—0.95。【结论】新疆霍城大西沟的野杏具有丰富的遗传多样性,遗传多态性比率达到90%以上,引物平均多样性信息指数(PIC)较高,接近1。61份野杏单株间的遗传相似度较高,亲缘关系较近。[ Objective]Through the analysis of ISSR markers, this project aims to analyze the genetic diversity of Xinjiang wild apricot germplasm resources and provide theoretical basis for utilization and protection of the resources and technical support. [ Method] Using ISSR markers, analysis of 61 samples of Daxigou wild apricot plant of genetic diversity. [ Result] The selection of 21 primers for PCR amplification of samples, a total of 437 amplified bands, of which 408 bands were polymorphic, percentage of polymorphic bands was 92. 46%.
