摘 要:【目的】寻找IGFBP-3基因在中国美利奴羊(新疆型)中的SNP位点,分析基因的多态性,并分析不同基因型个体在体重、油汗、光泽、毛长度评分、毛弯曲评分、毛弯曲数、毛细度评分、体格大小、剪毛量、平均直径、变异系数等经济性状上的差异显著性,为羊毛性状的候选基因提供理论基础。【方法】通过PCR-SSCP的方法,检测分析基因的多态性,用Excel软件进行数据的初步整理,用SAS8.1软件进行方差分析。【结果】得到两种基因型:AA和AB两种基因型,基因型频率分别为0.78和0.22,等位基因A和B的基因频率分别为0.89和0.11,A等位基因为优势等位基因,序列分析表明突变发生在序列片段的81碱基处且为G→C突变(g.81 G>C)。所研究绵羊群体在该位点为Hardy-Weinberg不平衡状态(0.01<P<0.05)。【结论】不同基因型的个体间在毛长性状上差异极显著,AA基因型要低于AB基因型(P<0.01);在毛细度评分上差异显著(0.01<P<0.05);在体重、油汗、光泽、弯曲数、体格大小、剪毛量、平均直径和变异系数上差异不显著(P>0.05)。IGFBP-3基因可以作为毛长度选择的一个候选基因。【Objective】Looking for the SNP in Chinese Merino sheep( Xinjiang type),carrying out the analysis of gene polymorphism and the significance of difference that exists in different individuals of genotype within terms of weight,stickiness,sweat,Sweat,sheen,grade of length,grade of bending,NO. of grade of finenesfineness,body size,fleece weight,average diameter,variable coefficients such as ect. economic traits in order to provide theoretical basis for candidate gene about wool.【Method】Making the analysis of gene polymorphism by the method of PCR- SSCP test. Collating Preliminary preliminary data using Excel excel software and making the analysis of variance using SAS8. 1 software.【Result】We obtained from Ttwo genotypes were obtained detected by PCR- SSCP( AA genotype and AB genotype),the genotype frequencies were 0. 78 and0. 22. Allele frequency of A and B were 0. 89 and 0. 11. A allele was dominant one. Sequence analysis revealed that a G / C transversion took place at the position 81 of fragment. The analysis from the date showed that the sheep group in this locus was Hardy- Weinberg imbalance condition( 0. 01 P 〈0. 05).【Conclusion】The individuals of genotype AA,AB had no significant difference in weight,oily sweat,sheen,bending number,body size,average diameter,variable coefficient traits( P 〈0. 05); Significant difference existed in the wool length existing in different genotypes( P 〈0. 01),in which,the wool length of genotype AA individuals was significantly shorter than that in genotypee AB individuals( P 〈0. 01); There was difference in grade of fineness in different genotypes( 0. 01 〈P 〈0. 05). IGFBP- 3 could be used as a candidate gene related to the length of wool in sheep.
