摘 要:目的探讨白细胞介素13(IL-13)基因第4外显子区+2044G/A单核苷酸多态性与哮喘易感性的关系。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法对100例支气管哮喘患者和100例健康对照者IL-13基因+2044G/A位点单核苷酸多态性进行检测,分析其基因型和等位基因分布频率。结果对照组GG、GA、AA基因型频率分别为70.0%、25.0%和5.0%,哮喘组为51.0%、39.0%和10.0%(χ2=7.71,P<0.05)。A等位基因频率在哮喘组为29.5%,对照组为17.5%,差异有统计学意义(χ2=8.01,P<0.01)。经统计学分析,哮喘患者与健康对照者的基因型与等位基因频率差异有统计学意义(P<0.05)。结论 IL-13基因+2044位点G/A多态性与支气管哮喘的发病密切相关,A等位基因可能是哮喘的易感基因。Objective To explore the roles of the interleukin-13(IL-13)gene exon4+2044G/A single nucleotide polymorphism(SNP)in the pathogenesis of bronchial asthma.Method Total of 100 asthmatic patients and 100 healthy controls were genotyped by using polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)methods.Result The genotype frequency of GG,GA,AA was 70.0%,25.0%and 5.0%in controls,51.0%,39.0% and 10.0%in asthma respectively.A allele frequency was higher in asthma compared with normal controls(29.5% vs 17.5%,P〈0.05).The distribution of genotype and allele frequency in +2044G/A site of IL-13 gene between two groups that there were statistical significantly differences(P〈0.05).ConclusionIL-13gene+2044G/A mutation has obvious relative with asthma.Allele may be the susceptible gene in the asthma.
