摘  要:目的探讨雌激素对体外培养的人子宫内膜癌(endometrial carcinoma,EC)JEC细胞中p57^(kip2)和Cyclin E表达的影响。方法体外培养的JEC细胞分别用高、中、低三种浓度(10^(-6)、10^(-8)、10^(-10)mol/L)的雌二醇(β-Estradiol,E2)治疗处理,于作用24、48、72 h后,分别用倒置显微镜和透射电镜观察JEC细胞的形态变化,用MTT、流式细胞术(flow cytometry,FCM)检测细胞的生长情况和细胞周期分布的变化,用Western blot法检测p57^(kip2)、Cyclin E蛋白的表达。结果形态观察、MTT、FCM检测显示,在中、低浓度的E2对JEC细胞的生长有促进作用(P<0.05);E2作用72 h后,处于G0/G1期细胞明显减少,S期细胞明显增多(P<0.05)。Western blot结果显示,E2作用于JEC细胞:(1)24 h时,p57^(kip2)蛋白在中浓度组表达最低,高浓度组表达最高(P<0.05),Cyclin E蛋白在高浓度组表达最低,中浓度组表达最高(P<0.05),然而,在48 h时,随着浓度的升高,p57^(kip2)、Cyclin E蛋白的表达均逐渐增加(P<0.05),72 h亦然。(2)p57^(kip2)蛋白在中浓度组随着作用时间的延长,表达增高(P<0.05),高浓度组亦然;Cyclin E蛋白在中浓度组随着时间的延长,表达降低(P<0.05),低浓度组亦然。p57^(kip2)、Cyclin E蛋白表达之间无相关性(P>0.05)。结论 p57^(kip2)、Cyclin E蛋白在EC发生、发展中的作用均受到E2的影响。随着E2作用浓度增加、作用时间延长,JEC细胞由增殖促进状态变成增殖抑制状态,中、高浓度组p57^(kip2)蛋白表达逐渐增高;中、低浓度组则随着E2作用时间的延长,Cyclin E蛋白表达逐渐降低。Purpose To investigate the effects of estrogen on the expressions of p57^kip2 and Cyclin E in human endometrial carcinoma (EC) cells named JEC in vitro. Methods The JEC cells cultured in vitro were treated with high, medium and low concentrations of 13-Estrogen (10^-6, 10^-8, 10^-10 mol/L), after 24, 48, 72 hours, the cells were observed on morphological changes by light and electron microscope. MTF and flow cytometry (FCM) were used to test the growth state and the cell cycle distribution changes of JEC ceils. Western blot was used to test the expressions of p57^kip2 and Cyclin E proteins in JEC cells. Results Morphology observation, MTT, FCM results showed when β-Estrogen was in medium and low concentration range, it was promoting JEC cells' growth (P 〈 0. 05 ). β-Estrogen obviously increased the cell percentage at the S phase of cell cycle and decreased at the G0/G1 phase in 72 hours (P 〈 0. 05 ). Western blot results showed, the expression of p57kip2 protein is the lowest in the medium concentration group, and the highest in the high concentration group ( P 〈 0. 05 ), the expression of Cyclin E protein is the lowest in the high concentration group, and the highest in the medium concentration group (P 〈 0.05 ) at 24 hours, the expressions of p57kip2 and Cyclin E proteins increased along with the increase of the concentration at 48 hours ( P 〈 0. 05 ), as well as 72 hours. In the medium concentration group, the expression of p57kip2 protein increased along with the time passed (P 〈 0. 05 ) , as well as in the high concentration group, in the medium concentration group, the expression of Cyelin E protein was decreased along with the time passed ( P 〈 0. 05 ) , as well as in the low concentration group. There was no correlation between the expressions of p57kip2 and Cyclin E proteins (P 〉 0. 05 ).
