摘 要:目的 探讨巨噬细胞移动抑制因子(MIF)和白细胞分化抗原74 (CD74)在妊娠期急性胰腺炎大鼠子代肝脏中的表达及意义.方法 妊娠晚期SD大鼠24只,随机分为假手术组(Sham)和胰腺炎组(AP).AP组经胆胰管逆行注射5%牛磺胆酸钠制模,Sham组则注入等量生理盐水,分3、6、12h3个时间点.术后在对应时间点分批剖杀.取各组孕鼠胰腺及胎鼠肝脏行苏木素-伊红(HE)染色光镜观察病理改变并评分;检测各组孕鼠血清中血清淀粉酶(AMY)、丙氨酸转氨酶(ALT)、天门冬氨酸转氨酶(AST)的表达;免疫组织化学法检测各组胎鼠肝脏MIF和CD74的表达.结果 AP组孕鼠胰腺病理评分(3、6、12 h各时点分别为4.83±1.12、10.50±1.03、11.78±1.25)及胎鼠肝脏的损伤(3、6、12 h各时点分别为1.33±0.50、1.56±0.53、2.33±0.71)及随着病程延长均逐渐加重(P<0.05),AP组血清AMY(3、6、12h各时点分别为3926.61±156.20、8 621.20±183.72、10273.45±325.41),ALT(3、6、12 h各时点分别为138.64±67.39、345.70±34.41、359.25±74.89),AST(3、6、12h各时点分别为665.63±105.09、734.11±94.69、844.35±121.66)的水平随病程延长逐渐升高(P<0.05),胎肝中MIF(3、6、12 h各时点分别为1.33±0.50、1.56±0.53、2.33±0.71)及CD74(3、6、12h各时点分别为5.37±0.71、10.38±1.45、3.98±0.57)的表达(分别为)呈先升高后降低的时间依赖性改变,6h时表达最高(P<0.05).结论 MIF和CD74在妊娠期急性胰腺炎大鼠子代肝脏中表达,可能在肝脏损伤的发生、发展过程中发挥重要作用.Objective To investigate the expression of macrophage migration inhibitory factor (MIF) and cluster of differentiation 74 (CD74) in fetal liver of acute pancreatitis rat in pregnancy.Methods Twenty-four SD rats in later gestation (gestation days =15-17) were obtained and randomly divided into two groups : Sham group and acute pancreatitis (AP) group.Each group was subdivided into 3 time points: 3 h, 6 h and 12 h.5% sodium taurocholate (STC, 0.1 ml/body weight) was used to inject into biliopancreatic duct through duodenum retrogradely to induce acute pancreatitis model in AP group, isotonic saline was substituted for 5% STC as a control in Sham group.Maternal serum were collected to measure serum amylase (AMY), alanine aminotransferase (ALT) and aspartate aminotransferase (AST), maternal pancreas and fetal liver were harvested for histological scoring and grading, immunohistochemistry assay was used to detect MIF and CD74 expressions in fetal liver (P 〈 0.05) in each time point.Results Maternal pancreas (histological scoring of pancreatitis at 3, 6, 12 hours was 4.83 ± 1.12, 10.50 ± 1.03, 11.78 ± 1.25 respectively) and fetal liver injury (histological grading at 3, 6, 12 hours was 1.33 ±0.50, 1.56 ±0.53, 2.33 ± 0.71 respectively) are gradually increased in AP group (P 〈 0.05), serum AMY (serum AMY at 3, 6, 12 hours after AP was 3 926.61 ± 156.20, 8 621.20± 183.72, 10273.45± 325.41 respectively), ALT (serum ALT level at 3, 6, 12 hours after AP was 138.64 ±67.39, 345.70 ± 34.41, 359.25 ±74.89 respectively) and AST (serum AST at 3, 6, 12 hours after AP was 665.63 ± 105.09, 734.11 ± 94.69,844.35 ± 121.66 respectively) are increased in the same pattern of organ injury in AP group (P 〈 0.05), MIF (MIF level at 3, 6, 12 hours after AP was 1.33 ± 0.50, 1.56 ± 0.53,2.33 ±0.71 respectively) and CD74 (CD74 level at 3, 6, 12 hours after AP was 5.37 ±0.71, 10.38 ± 1.45, 3.98 ± 0.57 respectively) expression in AP group are sig
