摘  要:目的 观察CO2培养浓度对A549细胞活性,合成和分泌肺泡表面活性蛋白C(SP-C)的影响.方法 A549细胞按CO2培养浓度分为:A组(5%CO2);B组(10% CO2);C组(18% CO2).各组在CO2培养24、36、48 h后,流式细胞仪检测细胞周期和凋亡率;反转录-聚合酶链反应(RT-PCR)检测SP-C mRNA表达;酶联免疫吸附法检测细胞培养液中SP-C浓度.结果 (1)与A组比较,B组G1峰和凋亡率在24、36、48 h,差异无统计学意义(P>0.05);C组与B组比在24h(68.0±6.2比55.0±3.8,P<0.01)、36 h(71.0±2.0比63.0±1.3)出现G1峰升高(P<0.05),并于36 h(10.0±1.2比6.9±1.9)、48 h(12.8 ±2.6比7.3±2.0)凋亡增加(P<0.05).(2)3组SP-C mRNA表达在24h比较差异无统计学意义(P>0.05);与A组比较,B组在36 h(0.32±0.02比0.24±0.02)(P<0.01)、48 h(0.24±0.03比0.16±0.03)SP-C mRNA表达升高(P<0.05),而C组均降低(0.17 ±0.04、0.10 ±0.02) (P <0.05).(3) SP-C 3组浓度在24h比较差异无统计学意义(P>0.05);与A组比较,B组SP-C浓度在36 h(31.00±2.00比21.00±0.85)(P<0.01),48 h(42.00±1.02比26.00±0.97)升高(P<0.05);C组SP-C浓度在48 h(20.00±0.51比26.00±0.97)降低(P<0.05),24、36 h比较差异无统计学意义(P>0.05).结论 A549细胞活性和SP-C的表达受CO2培养浓度的影响;随着CO2浓度的升高,其作用由上调转为抑制.Objective To investigate the effects of CO2 concentration on the activity of A549 cell, the synthesis and secretion of puhmonary surfactant protein C (SP-C).Methods A549 cells in CO2 culture were divided into three groups, group A (5% CO2);B group (10% CO2);group C (18% CO2).After culture of 24, 36, 48 h.The cell cycle progression and apoptosis were measured by flow cytometric analysis, the expression of SP-C mRNA was measured by reverse transcriptase-polymerase chain reaction (RT-PCR), and the SP-C concentration in the cell culture supernatants was measured by enzyme-linked immunosorbent assay method.Results (1) Compared with A group, B group' s G1 peak and the rate of apoptosis in 24, 36 and 48 h showed no significant difference (P 〉0.05).In the C group, The G1 peak increase in 24h (68.0 ±6.2 vs.55.0 ±3.8) and 36 h (71.0 ±2.0 with 63.0 ± 1.3) (P 〈0.01;P 〈 0.05), and the apoptosis rate increased in 36 h (10.0 ± 1.2 vs.6.9 ± 1.9) and 48 h (12.8 ± 2.6 with 7.3 ± 2.0) (P 〈 0.05).(2) The SP-C mRNA expression of the three groups in 24 hours were no difference (P 〉 0.05).According to A group, B group' s mRNA of SP-C increased in 36 h (0.32 ± 0.02 vs.0.24±0.02) (P〈0.01) and48 h (0.24±0.03 vs.0.16±0.03) (P〈0.05);On the contrary,they are all drop in the C group (0.17 ±0.04, 0.10±0.02) (P〈0.05).(3)The SP-C concentration of the three groups in 24 hours were no difference (P 〉 0.05).According to A group, B group's concentration ofSP-C increased in 36 h (31.00 ±2.00 vs.21.00 ±0.85, P〈0.01) and 48 h (42.00 ± 1.02 vs.26.00 ± 0.97, P 〈 0.05);Compared with A group, C group' s concentration of SP-C decreased in 48 h (20.00 ± 0.51 vs.26.00 ± 0.97, P 〈 0.05), and 24 h and 36 h showed no significant difference (P 〉 0.05).Conclusion The activity of A549 cells and the expression of SP-C were influenced by the CO2 concentration of culture.With the increasing of CO2 concentrati
