摘 要:目的观察复方丹参液对大鼠缺血再灌注心肌的影响,探讨复方丹参在心肌缺血再灌注损伤过程中对心肌的保护机制。方法建立左冠状动脉前降支缺血再灌注大鼠动物模型。实验分组:A组为假手术对照组(n=20):仅游离大鼠冠状动脉前降支,不结扎;B组为缺血再灌注损伤组(n=20),缺血30min,再灌注60min;C组为复方丹参液处理组(n=20),于缺血前30min经腹腔注射复方丹参注射液15ml/kg,余同B组。再灌60min后,采用酶联免疫吸附实验(ELISA)测定3组血浆肿瘤坏死因子-α(TNF-α)、白介素-10(IL-10)浓度;应用TTC染色法评价3组心肌损伤程度;采用免疫印记(Western blot)检测心肌核因子-κB(NF-κB)蛋白活性水平。结果与A组比较,B组TNF-α、IL-10的浓度,心肌组织NF-κB蛋白含量明显增高(P<0.01);与B组比较,C组梗死面积明显减少(P<0.01);TNF-α、IL-10浓度浓度降低,心肌组织NF-κB蛋白含量减少(P<0.01)。结论复方丹参对大鼠心肌缺血再灌注损伤有保护作用,可以减少心肌梗死面积,其可能机制与抑制NF-κB蛋白表达,减少炎性因子TNF-α、IL-10等释放有关。Objective To explore the protective effect of Danshen Injection on myocardial ischemia and reperfusion (I/R) injury in rats and its mechanism. Methods 60 male SD rats were randomly divided into three groups : Group A ( n = 20) : the anterior descending branch of left coronary artery was exposed and a piece of silk thread was placed around the artery but not tied. Group B (n = 20) :the left coronary artery was ligated for 30 min, then followed by reperfusion for 60 rain. Group C (n = 20) :Danshen (1.5 ml/kg)was injected in- traperitoneally at 30 minbefore ischemia. After reperfusion, the blood samples were taken and the plasma levels of tumor necrosis faetor-a (TNF-a)and Interleukin-10(IL-10) were measured with enzyme-linked immunosor- bent assay (ELISA) method. The myoeardium was stained with TI'C to evaluate the infarction volume. Western blotting technique was used to detect the expression of myocardial Nuclear Factor-kappa B (NF-KB). Results Compared with group A, the contents of TNF-cx and IL-10, as well as the expression of NF-KB in myocardial tis- sue of group B was significantly elevated (P 〈 0.01 ). Compared with group B, Danshen injection could obvious- ly decrease the Infarction volumeand the contents of TNF-cx andlL-10, reduce the expression of NF-KB ( P 〈 0. 01 ). Conclusion Danshen injectioncan reduce myocardial ischemia-reperfusion injury in rats ,which might be related to the inhibition of the activty of NF-KB and down-regulation of TNF-a and IL-10 production.
