摘 要:目的 :检测一种染发剂的致突变性作为毒理学评价的一部分。方法 :用Ames试验和体外哺乳动物细胞 (CHL)染色体畸变试验来检测氧化型染发剂的致突变性。结果 :在加S9mix条件下 ,每皿 2 5 0 0 μg至 5 0 0 0 μg对测试菌株TA98具有明显的诱变性 ,其回变菌落数是自发回变数的 2~ 3倍 ,Ames试验检测结果为阳性。体外哺乳动物细胞 (CHL)染色体畸变试验显示在加或不加S9mix条件下 ,当受试物终浓度为 30 0 μg/ml,6 0 0 μg/ml和 10 0 0 μg/ml,除了 30 0 μg/ml在加S9mix时染色体畸变细胞率未显著增加外 (P >0 0 5 ) ,其余浓度无论加或不加S9mix,染色体畸变细胞率与阴性对照组相比均有显著性差异 (P <0 0 5 )。 结论 :受试的氧化型染发剂具有一定的遗传毒性。Purpose: To study the mutagenicity for a hair dye as a part of toxicological evaluation. Methods: The Ames test and chromosome aberration test in vitro in CHL cells were used. Results: At the dose of 2 500 μg/plate to 5 000 μg/plate with the S-{9mix}, the sample can induce the mutation of the strain TA98, the number of reverse mutation bacteria clone was 2~3 times higher than the number of spontaneous mutation bacteria clone. CHL cell chromosome aberration test showed that at the concentration of 300 μg/ml without the S-{9mix}, and at concentration of 600 μg /ml and 1 000 μg/ml with or without the S-{9mix}, the sample can induce the chromosome aberration. The rate of chromosome aberration in treated group was significantly higher than that in the negative control group. Conclusion: The oxidative hair dye tested has certain genetic toxicity.
