宣肺平喘胶囊对慢性阻塞性肺疾病大鼠模型肺组织的病理形态学影响

摘　　要：目的:研究宣肺平喘胶囊对慢性阻塞性肺疾病(COPD)的病理形态学的影响。方法:采用香烟烟熏联合气管内注入脂多糖方法建立COPD大鼠模型。造模成功后,随机分为宣肺平喘胶囊低、中、高剂量组,模型组以及对照组。空白组和模型组以蒸馏水灌胃,对照组以地塞米松混悬液灌胃;其余治疗组以宣肺平喘颗粒混悬液灌胃,时间30天,每天1次。末次灌胃后,观察大鼠一般情况(如咳喘、毛发、体重、饮食、活动等);对左下肺进行HE染色,病理组织观察。结果:一般情况,除空白组外,均表现为病态,但阳性对照组和中药高剂量组之间表现接近,中剂量组和低剂量组表现更差;肺组织病理改变也是阳性对照组和中药高剂量组病变较轻且两组病理改变接近,而中剂量组和低剂量病理改变呈梯度加重,提示中药对肺组组织病理学的影响与剂量呈正相关。结论:宣肺平喘胶囊能够改善大鼠一般情况;光镜下所见,能够使支气管腔内分泌物减少,支气管纤毛脱落减轻,且改善情况与剂量呈正相关。Objective ： To study the effects of xuanfei pingchuan capsules on the pathomorphism of model rats with chronic obstructive pulmonary diseases . Methods：The models of COPD rats were established by passive cigarettesmoking and intratracheal LPS instillation. After the models were established successfully, COPD rats were randomly divided into xuanfei pingchuan capsules group in low, medium and high dose , blank group and control group. The blank group and the model group were given distilled water by gavage. The control group was given dexamethasone suspension by gavage. The rest of the treatment groups were treated with xuanfei pingchuan granules suspension by gavage. All were treated once daily for 30 days . After the last gavage, general conditions as cough and dyspnea, hair, weight, diet and activity were observed. Then stained the left lower lobe with hematoxylin and eosin, and observed its biopsy. Results： Under light microscope, the endobronchial secretions were reduced, and the sloughing of the bronchial cilia were reduced. Conclusion ： Xuanfei Pingchuan capsules can improve the general conditions of COPD rats. The mechanisms could include reducing the formation and release of the proinflammatory mediators and reducing the secretion of mucus in the airway.
