摘  要:目的探讨60Coγ射线亚致死量辐射对小鼠外周血micro RNA(miRNA)的影响及意义。方法无特定病原体(specific pathogen free,SPF)级C57BL/6J小鼠接受2 Gy全身照射后,分别于照射后6、24和72 h进行外周血白细胞计数,同时用Agilent miRNA芯片技术筛选小鼠外周血差异表达的miRNA。结果照射后6 h差异表达miRNA共16个,其中13个上调,3个下调;照射后24 h差异表达miRNA共36个,其中13个上调,23个下调;照射后72 h差异表达miRNA共30个,其中13个上调,17个下调。且与未照射组比较差异表达miRNA有统计学意义(P<0.05)。筛选15个miRNA组成的表达谱,对辐射损伤时间的判断有很好的效果,准确率在90%以上。2 Gy照射后,24 h和72 h外周血白细胞总数与未照射组比较,差异有统计学意义(P<0.05)。miRNA表达量化方面优于血液中白细胞的变化。结论外周血miRNA差异表达与辐射损伤有关,为探索早期辐射损伤后的辅助诊断指标提供了一定参考。Objective To investigate the differential expression profiles of microRNA(miRNA) induced by sublethal dose of radiation on mouse peripheral blood and to explore their significances. Methods After specific pathogen free ( SPF ) degree C57BL/6J mice exposed to 2 Gy whole body radiation, total number of peripheral white blood cell count were measured at 6 h, 24 h and 72 h. Changes of miRNA expression in peripheral blood were analyzed by using Agilent miRNA microarrays 6 h, 24 h and 72 h after radiation. Results 6 h after 2 Gy 60Coy -irradiation, miRNA microarray revealed that 16 miRNA were differentially expressed, in which 13 miRNA upregulated, 3 miRNA down-regulated. 36 miRNA and 30 miRNA were differentially expressed respectively at 24 h and 72 h after radiation. Compared with control group, the total number of peripheral white blood cell count at 24 h and 72 h decreased obviously after 2 Gy whole body irradiation ( P〈0.05 ). Conclusions miRNA were involved in the regulation of radiation damage after sublethal dose, and miRNA expression change derived from mouse blood may be used as new biomarkers for exposure to sublethal dose radiation.
