摘 要:目的:观察温阳益气方对脾肾阳虚型溃疡性结肠炎(UC)大鼠模型Th17细胞的影响。方法:40只SPF级Wistar大鼠运用TNBS造模法,成功制备出脾肾阳虚型UC大鼠模型后,随机分为4组:正常组、模型组、温阳益气(WYYQ)组、美沙拉嗪(MT)组。药物干预28 d后取标本。运用HE染色法进行UC大鼠结肠黏膜组织病理学观察,采用酶联免疫吸附试验(ELISA)检测大鼠血清中IL-17、IL-23的表达;采用实时荧光定量PCR法定量分析结肠组织中RORγt mRNA的表达;采用Western Blot法测定结肠组织中Stat3蛋白的表达。结果:病理切片HE染色后在光镜下观察,可见结肠黏膜水肿、糜烂、出血,黏膜上皮缺损,溃疡灶形成。与模型组相比,WYYQ组和MT组镜下病理组织评分明显降低(P<0.05),血清IL-17、IL-23含量明显降低(P<0.05),结肠组织中RORγt mRNA的表达明显降低(P<0.05),Stat3蛋白的表达明显降低(P<0.05)。结论:温阳益气方能明显改善UC模型大鼠的炎症情况和黏膜组织的病理情况,降低血清IL-17、IL-23的含量,降低结肠组织中RORγtmRNA和Stat3的表达。抑制和降低UC模型大鼠结肠组织中Th17细胞的分化,可能是温阳益气方发挥其治疗溃疡性结肠炎的作用机制之一。Objective : To explore the pathogenesis of yang deficiency in the UC occurrence, and observe the influence of method of warming yang and nourishing qi on Th17 cells of UC model rats with Syndrome of Spleen - Kidney Yang Deficiency. Methods: First, UC model rats with Syndrome of Spleen - Kidney Yang Deficiency were established by using TNBS method; Then the rats were randomly divided into the following 4 groups: the normal group, the model group, the warming yang and nourishing qi (WYYQ) grnup, and MT group. After 28 days'medicine intervention, the specimen were taken out. The colonic mucosa pathol- ogy of UC rats were observed by HE staining method. The serum IL- 17 and IL -23 expressions were detected by ELISA. The RORγt mRNA expression in colon was analyzed by PCR method. The Stat 3 protein expression in colon was measured by Western Blot method. Results: After pathological section was colored by HE staining method, then observed under the light microscope, colonic mucosal edema, erosion, bleeding, mucous membrane epithelial defects, ulcer lesions could be seen. Compared with the model group, pathological tissues scores of the WYYQ group and the MT group decreased obviously (P 〈0.05) ; Serum IL- 17 and IL - 23 contents decreased obviously( P 〈 0.05 ) ; ROR-γt mRNA expression of colon decreased obviously( P 〈 0.05 ) ; Star 3 protein expression decreased obviously (P 〈 0.05 ). Conclusion : WYYQ can obviously improve the inflammation of UC model rats, effectively enhance colonic mucosa pathology of UC rats, reduce serum IL - 17 and IL - 23 contents, the expressions of RORTtmRNA and Stat3 of colon, impress and reduce Thl7 cells differentiation, which probably is one of the mechanisms of WYYQ method in treating UC.
