摘 要:目的观察冠心舒通胶囊对心肌梗死后慢性心力衰竭大鼠心肌能量代谢的影响及可能机制。方法 80只SD大鼠随机选取20只作为空白组,余采用左冠状动脉前降支结扎法配合力竭式游泳及节食方法复制慢性心力衰竭大鼠模型。将造模成功的大鼠随机分为模型组、西药组、冠心舒通胶囊组。造模后西药组给予赖诺普利片1.5 mg/(kg·d)灌胃,冠心舒通胶囊组给予冠心舒通胶囊10.0 g/(kg·d)灌胃,空白组和模型组分别给予等容积蒸馏水3 ml/次灌胃,均每日1次。持续28天后,测定各组大鼠血浆B型尿钠肽(BNP)、三磷酸腺苷(ATP)浓度,测定心肌组织过氧化物酶体增殖活化受体γ辅助活化因子1α(PGC-1α)、核呼吸因子1(NRF-1)、线粒体转录因子A(mt TFA)基因和蛋白表达。结果与空白组比较,模型组、西药组、冠心舒通胶囊组血浆BNP水平升高,血浆ATP浓度和心肌组织PGC-1α、NRF-1、mt TFA mRNA及蛋白表达降低(P<0.05)。与模型组比较,西药组、冠心舒通胶囊组血浆BNP水平降低,而ATP浓度、PGC-1α、NRF-1、mt TFA mRNA及蛋白表达均升高(P<0.05),并且西药组、冠心舒通胶囊组组间比较差异无统计学意义(P>0.05)。结论冠心舒通胶囊可能通过激活与心肌能量代谢有关的PGC-1α、NRF-1、mt TFA因子,增加线粒体的产能,从而改善和纠正心力衰竭。Objective To observe effect and possible mechanism of Guanxin Shutong Jiaonang( 冠心舒通胶囊)on myocardial energy metabolism of chronic heart failure model rats after myocardial infarction. Methods Twenty of eighty SD rats were randomly selected as the blank group,and the others were copied to rats model with chronic heart failure used ligation of left coronary artery anterior descending branch cooperated with exhausting swimming and diet. Rats with successful model were randomly divided into model group,western medicine group and Guanxin Shutong Jiaonang group. The western medicine group was given lisinopril 1. 5 mg /( kg·d) to gavage. The Guanxin Shutong Jiaonang group was given Guanxin Shutong Jiaonang 10. 0 g /( kg·d) to gavage. The blank group and the model group were respectively given distilled water 3 ml each time to gavage,once a day. After 28 days,the researchers measured plasma b-type natriuretic peptide( BNP),triphosadenine( ATP),gene and protein expression of peroxisome proliferator-activated receptor γ auxiliary activation factor 1α( PGC-1α),nuclear respiratory factor 1( NRF)and mitochondrial transcription factor A( mt TFA) of myocardial tissue in rats of each group. Results Compared with the blank group,plasma BNP level increased in the model group,the western medicine group and the GuanxinShutong Jiaonang group but plasma ATP concentration and PGC-1α,NRF-1,mt TFA mRNA and protein expression of myocardial tissue decreased( P〈 0. 05). Compared with the model group,plasma BNP level of the western medicine group and the Guanxin Shutong Jiaonang group decreased but ATP concentration,PGC-1α,NRF-1,mt TFA mRNA and protein expression increased( P〈 0. 05),and the inter-group difference had no statistical significance between the western medicine group and the Guanxin Shutong Jiaonang group( P〉 0. 05). Conclusion Guanxin Shutong Jiaonang may improve and correct heart failure by activating PGC-1α,NRF-1,mt TFA factor associated with myocardial ene
