重组人卵泡刺激素类似物对幼龄小鼠卵巢发育的影响

摘　　要：目的:探讨重组人卵泡刺激素(recombinant follicle stimulating hormone,r FSH)类似物对幼龄小鼠卵巢发育的影响。方法:3周龄小鼠随机分为6组:溶媒空白对照组、果纳芬单次注射组(0.5 IU)和果纳芬3次注射组(0.5 IU·d-1,连续给药3 d);r FSH类似物低剂量组(0.375 IU)、中剂量组(0.75 IU)和高剂量组(1.5 IU)均在停药24 h后摘取卵巢称重并计算卵巢指数,卵巢组织经HE染色,观察各级卵泡所占比例。结果:与空白对照组相比,r FSH类似物能剂量依赖性地促进卵巢增重,显著增加卵巢指数(P<0.05)。0.375 IU剂量即可显著促进幼龄小鼠卵巢指数的增加(P<0.05),1.5 IU剂量组较阳性药物果纳芬3次注射组无显著性差异;0.75 IU r FSH类似物较空白对照组可显著降低原始卵泡所占比例,提高成熟卵泡和黄体所占比例(P<0.01),且1.5 IU剂量组较阳性药物果纳芬3次注射组能更有效地降低原始卵泡的比例,提高成熟卵泡和黄体所占比例(P<0.01)。结论:r FSH类似物不仅能够促进幼龄小鼠卵巢增重以及促进原始卵泡、初级卵泡和次级卵泡的发育,而且具长效性,即单次注射即可使血药浓度维持3 d。Objective ： To investigate the effect of recombinant follicle stimulating hormone （rFSH） analog on development of ovary in immature mice. Methods： Three-week-old mice were randomly divided into 6 groups, and treated with solvent （control） , single injection of Gonal-F （0.5 IU）, 3 day-injection of Gonal-F （0.5 IU·d^-1 ） and single injection of long-acting rFSH analog （0. 375, 0. 75 and 1.5 IU）. All the mice were killed at 24 h after the last injection, and the ovaries were removed and weighed. Then, the organ index was calculated, and the ratios of different stages of follicles were analyzed after HE staining. Results ： Single injection of rFSH analog effec- tively increased the ovarian weight and organ index of immature mice in a dose-dependent manner （P 〈 0. 05 ） com- pared to the control group. It obviously increased the ovarian organ index at a dose of 0. 375 IU （P 〈 0. 05 ） , but multiple injection of Gonal-F （ 1.5 IU for 3 days） had no significant effect. Moreover, rFSH analog （0.75 IU） re- duced the proportion of the primordial follicle, and increased the ratios of mature follicle and luteum （P 〈 0.01 ）. It was redUced the ratio of primordial follicle and increased the ratio of mature follicle and luteum more effectively than Gonal-F 3 （1.5 IU for 3 days, P 〈0. 01 ）. Conclusion： rFSH analog can effectively induce ovarian growth and multiple follicular development with a long-acting property. Single injection of this analog has the similar effica- cy with consecutive administration （3 days） of the commercially available rFSH preparation Gonal-F.
