山东汉族TIM-1基因多态性与2型糖尿病肾脏病的相关性

摘  要:目的探讨T细胞免疫球蛋白域和黏蛋白域蛋白-1(TIM-1)基因多态性与山东汉族2型糖尿病肾病的相关性。方法采用病例-对照的方法,收集2型糖尿病肾病患者112例,正常对照组201例。采用改良的聚合酶链反应-限制性片段长度多态性(PCR-RFLP)检测TIM-1基因的两个SNP位点:rs1039438和rs6878732,计算其基因型频率及等位基因频率并统计分析。结果 rs1039438位点的基因型频率与等位基因频率在对照组和2型糖尿病肾病组中有显著的统计学差异(P值分别为0.005和0.002),rs6878732位点的基因型频率与等位基因频率在两组间无统计学差异。结论 TIM-1基因与山东汉族人群2型糖尿病肾病的发病有明显相关性,rs1039438位点的G等位基因可以增加2型糖尿病肾病发病风险,而此位点的A等位基因则为保护因素。Objective To investigate the association between TIM-1 gene polymorphism and the susceptibility of type 2 diabetic kidney disease in a Han population of Shandong Province. Methods In this case-control study, !.12 cases of type 2 diabetic kidney disease patients and 201 normal controls were collected. The two SNPs of TIM-1 gene:rs1039438 and rs6878732 in both groups were analyzed. Improved PCR-RFLP by their genotype frequency and allele frequency were calculated and compared. Results A significant difference between the DKD and control groups was found in the rs1039438 genotype and allele,no significant difference was found in rs6878732. Conclusion The rs1039438 SNP polymorphism of TIM-1 gene might be associated with susceptibility of type 2 diabetic kidney disease, allele G could increase the risk of this disease,while allele A might be a protective factor.
