摘  要:目的:研究创伤后应激障碍(PTSD)大鼠肠系膜和软脑膜微循环的改变及其机制。方法:成年健康雄性SD大鼠18只,随机分为正常对照组(n=6)和PTSD组(n=12)。采用单程延长刺激法复制PTSD大鼠模型,通过旷场实验检测大鼠的行为学改变;采用活体微循环观测技术观察大鼠肠系膜及软脑膜微血管内径和血流速度的改变;采用ELISA检测大鼠血浆去甲肾上腺素(NA)、内皮型一氧化氮合酶(eNOS)和内皮素-1(ET-1)水平。结果:与正常对照组比较,PTSD组大鼠在旷场中穿行格数及直立次数显著减少(P<0.01);肠系膜及软脑膜细动脉、细静脉管径明显变细(P<0.05),血流速度趋缓;血浆NA和ET-1含量显著升高(P<0.01),但eNOS水平无显著变化(P>0.05)。结论:PTSD大鼠存在肠系膜和软脑膜微循环障碍。Objective:To investigate the effect of post-traumatic stress disorder (PTSD)on microcirculation of mesentery and cerebral pia mater in rats and the underlying mechanisms.Method:Sprague-Dawley (SD)rats were randomly divided into two groups:control group (n=6)and PTSD group (n=12).The PTSD model was rep-licated by using the single prolonged stress method.On the 14th day post-stress,the changes of animals&#39;behavior was evaluated via open-field;the microcirculation of mesentery and cerebral pia mater of rats was observed using the microscope;the plasma noradrenaline (NA),endothelial nitric oxide synthase (eNOS)and endothelin-1 (ET-1 ) were measured by using enzyme linked immunosorbent assay (ELISA).Results:Compared with the control rats,the numbers of crossing and rearing of the PTSD rats were decreased significantly in the open-field (P 〈0.01);the di-ameters of arteriole and veinule in the mesentery and cerebral pia mater from the PTSD rats were significantly thin-ner (P 〈0.05),the blood flow velocity showed the trend of slowed down;the plasma levels of NA and ET-1 were elevated significantly (P 〈0.01);there was no significant difference in the plasma level of eNOS (P 〉0.05).Con-clusion:PTSD induces microcirculation dysfunction in mesentery and cerebral pia mater in rats.
