摘 要:目的:探讨辅助型T细胞9(Th9)对动脉粥样硬化斑块形成的影响。方法:8周龄载脂蛋白E基因敲除(apoE-/-)小鼠28只,4只用作制备Th9细胞,余24只分为四组:高脂喂养至12周龄组和16周龄组,Th9过继组(尾静脉注射Th9)和白细胞介素-9抗体(IL-9 Ab)组(尾静脉注射IL-9 Ab),每组6只。分别用流式细胞术及ELISA计数12周龄组和16周龄组小鼠外周血Th9细胞和检测IL-9水平变化,同时HE染色观察两组腹主动脉斑块面积及其与管腔面积的比值;16周时用相同方法检测Th9过继组和IL-9Ab组上述指标,并行两组腹主动脉CD68免疫组化(SP法)染色,比较CD68阳性细胞数量差异。结果:16周龄组小鼠外周血Th9细胞计数及IL-9水平均较12周龄组升高,腹主动脉斑块面积和斑块面积/管腔面积比值也增大,差异均有统计学意义(P<0.05);与IL-9Ab组比较,Th9过继组外周血Th9细胞数量及IL-9浓度均上升,腹主动脉斑块面积和斑块面积/管腔面积比值明显增大,斑块内CD68阳性细胞增多,纤维帽厚度变薄,差异均有统计学意义(P<0.05)。结论:Th9细胞可能通过IL-9促进斑块形成,并可能是斑块不稳定的重要因素。Objective:To explore the effect of Helper T cells 9 (Th9 )on the formation of atherosclerosis plaque.Method:28 apolipoprotein E gene knockout (apoE-/-)mice of 8 weeks of age were fed with high fat diet.4 mice was used for preparation of Th9 cells,the rest 24 were divided into four groups:12 weeks of age group and 1 6 weeks of age group and Th9 adoptive group (injection of Th9 via tail vein)and IL-9 Ab group (injection of IL-9 Ab via tail vein).Each group contained 6.Flow cytometry and ELISA assay were used to detect the changes of Th9 cell count and IL-9 level in peripheral blood of 12 and 1 6 week old mice,and HE staining was used to observe the area of abdominal aortic plaque and the ratio of the size of the abdominal aorta to the lumen area.The same method was used to detect the above indexes of Th9 and IL-9 Ab groups in 1 6 weeks.CD68 immunohistochemical (SP)staining was performed to compare the number of CD68 positive cells in the two groups.Results:The peripheral blood Th9 cells and IL-9 levels of 1 6 weeks old apoE-/- mice were higher than those of the 12 week old mice.The area of ab-dominal aortic plaque and plaque area/lumen area ratio also increased.The differences were statistically significant (P 〈0.05);comparison of Th9 adoptive group and IL-9 antibody group,Th9 cells quantity in peripheral blood and IL-9 concentration of the former increased,abdominal aortic plaque area and plaque area/lumen area ratio increased significantly,CD68 positive cells increased in the plaque,the thickness of fibrous cap became thin,the differences were statistically significant(P 〈0.05).Conclusion:Th9 cell could boost atherosclerosis and contribute to unsteady state of artery plaque via IL-9.
