摘 要:目的探讨乙酰唑胺对胸膜肿瘤小鼠AQP1表达及胸腔积液的影响。方法建立胸膜肿瘤动物模型,随机分为肿瘤组和干预组,干预组每日一次经口灌注乙酰唑胺(40 mg·kg-1·d-1)。比较各组小鼠胸腔积液的量,采用荧光定量PCR和免疫组化的方法测定水通道蛋白1及其mRNA的表达。结果胸膜肿瘤小鼠胸腔内见血性积液及肿瘤结节生长,干预组小鼠胸水量较肿瘤组减少,免疫组织化学染色显示AQP1表达于胸膜壁层上皮,干预组小鼠壁层胸膜AQP1水平较肿瘤组降低。结论乙酰唑胺可以抑制胸膜肿瘤小鼠壁层胸膜AQP1及其mRNA的表达,减少恶性胸腔积液。Objective To investigate the effect of acetazolamide to AQP1 and malignant plural effusion in mice with malignant pleuroma. Methods Pleura1 tumor animal models are built and mice were assigned into pleuroma group (Group A)and intervention group( Group B ). Mice in Group B were fed with acetazolamide (40 mg .kg- 1 . d- 1 ) dai- ly. Plural effusion of mice in these two groups were compared. The levels of AQP1 and AQP1 - mRNA were measured by immunohistochemistry and Real - Time PCR. Results The hemorrhagic effusion and tumor nodules in mice of group A and B, and the volume of MPE in group B decreased compared with that of group A, so did the expression of AQP1. Conclusion Acetazolamide can inhibit the generation of AQP1 and its mRNA's expression and reduce the volume of malignant pleural effusion.
