摘 要:目的通过研究急性冠脉综合征(ACS)患者外周血CD4+T细胞CD70基因启动子调节序列的甲基化状态,探讨ACS患者CD4+T细胞DNA甲基化异常在CD70表达中的作用。方法采用聚合酶链式反应(PCR)技术检测CD70mRNA表达水平;亚硫酸氢盐测序检测CD70(刑耶刀)基因启动子调节序列的甲基化状态。结果与正常对照组相比,CD70mRNA表达水平显著增加[(0.227±0.052)比(0.151±0.027)],CD70基因启动子甲基化水平显著降低,差异有统计学意义(P〈0.05)。CD70基因启动子DNA甲基化水平低于对照组(15.38±7.20)%比(27.18±3.27)%与。CD70mRNA表达水平呈显著负相关(r=-0.887,P=0.01)。结论ACS患者CD4+T细胞CD70基因启动子区域甲基化水平减低,并与CD4+T细胞CD70mRNA表达水平呈负相关,证实了ACS患者CD4+T细胞DNA甲基化异常导致ACS患者CD4+T细胞CD70异常表达。Objective To investigate the methylation of CD70(TNFSF7) promoter in CD4+T cells in patients with acute coronary syndrome (ACS). Methods The levels of CD70 mRNA were determined by PCR. The methylation status of CDT0 promoter regulatory sequences was detected by bisulfite sequencing. Results The levels of CD70 mRNA in CD4+ T cells was significantly higher in patients with ACS (P 〈 0.05 ). The meth- ylation status of CD70 gene promoter region was significantly lower than the normal control group (P 〈 0.05). We found a negative correlation between TNFSF7 promoter methylation and CD70 mRNA. Conclusion The CD70 gene promoter region was demethylation in CD4 + T cells from patient with ACS, and we also found a nega- tive correlation between TNFSF7 promoter methylation and CD70 mRNA levels. These finding suggest that dem- ethylation of CD70 gene promoter region contributes to the overexpression of CD70 in CD4 + T cells from patients with ACS.
