摘  要:根据GenBank收录的鸡SPRN(ChSPRN)序列设计特异性引物,以鸡脑组织基因组DNA为模板,无菌采集10只青脚麻鸡脑组织,克隆鸡朊样蛋白Shadoo的基因(SPRN)并分析其序列与高级结构特征,并应用生物信息学方法与软件进行分析,探讨其与PrP(朊蛋白)之间的相互关系.结果表明:ChSPRN长354bp,在第23位与第56位发生G→A、C→T突变,分别对应氨基酸序列C→Y、P→S突变,与GenBank中收录的鸡SPRN序列相比,两者核苷酸序列同源性高达99.7%,推导氨基酸序列同源性为99.9%.分析氨基酸序列并利用同源法构建ChSho与ChPrP的三级结构,结果显示它们N-端同源性为44.25%,C-端结构相似性为23.33%;ChSho与ChPrP在核苷酸、RNA二级结构、氨基酸水平以及高级结构中表现出的惊人相似性,推测其与PrP具有相似功能.The relationship between prion-like protein Shadoo gene(SPRN)and PrP(prion protein)of chicken was explored by cloning the SPRN and analyzing its amino acid sequence and advanced structural characteristics.According to the reported chicken SPRN(ChSPRN)sequence in GenBank,specific primers were designed to amplify the SPRN complete open reading frame(ORF)by using chicken brain tissue genomic DNA as template,furthermore,the SPRN was analyzed by bioinformatics methods and software.The results showed that the ChSPRN was 354 bp long,and there were G→A and C→T base mutation at 23 bp and 56 bp,respectively,corresponding to the amino acid sequence C→Y and P→S mutation.Compared with chicken SPRN sequence recorded in GenBank,their nucleotide sequence homology was up to 99.7%,the deduced amino acid sequence homology was 99.9%.After analyzing the amino acid sequence and building the tertiary structures of ChSho and ChPrP by the homologous method,we found that their N-end homology was 44.25%,and C-end structural similarity was 23.33%.ChSho and ChPrP showed striking similarity in nucleotide,RNA secondary structure,amino acid levels and advanced structure,thus we could speculatethat it had similar function with the PrP.
