摘  要:【目的】利用DNA条形码技术对新疆北部梭梭进行鉴定,从分子系统学角度探讨不同果翅颜色梭梭的亲缘关系。【方法】用通用引物ITS2对梭梭样品进行PCR扩增,得到的ITS2序列构建NJ系统进化树和遗传距离分析。【结果】ITS2引物的PCR扩增得到了500 bp左右的片段,最大K2P遗传距离为0.057,系统进化树结果显示,相同果翅颜色的梭梭能够聚为一支。【结论】ITS2序列能够很好的将不同果翅颜色的梭梭样品区分开,为梭梭的遗传多样性研究提供了一定基础。[ Objective ] This project aims identify Haloxylon in Northern Xinjiang using DNA bar code technology and investigate the genetic relationships of different colors of Haloxylon ammodendron fruit wings from the perspective of molecular system. [ Method ] Amplification of Haloxylon ammodendron samples with primers ITS2 for PCR, the ITS2 sequences were used to construct NJ phylogenetic tree and genetic distance analysis. [ Result] PCR was amplified by primer ITS2 to get about 500 bp fragment, the maximum K2P genetic distance was 0.057, the phylogenetic tree showed that the same color of Haloxylon fruit wings can be gathered into one. [ Conclusion ] ITS2 sequence could be very good to separate the different colors of Haloxylon ammodendron fruit wing sample, which might provide some basis to study the genetic diversity of Haloxylon ammodendron.
