益气活血化痰法对COPD模型大鼠细胞因子及气道重塑的干预作用

摘　　要：目的观察益气活血化痰法对慢性阻塞性肺疾病(COPD)模型大鼠细胞因子及气道重塑的影响,探讨益气活血化痰法治疗COPD的作用机理。方法通过香烟熏结合脂多糖(LPS)气管滴入法,建立COPD大鼠模型。观察益气活血化痰方药对大鼠血清中TNF-α、IL-8和肺泡灌洗液中MUC5AC的表达水平及其对各级支气管和肺组织病理变化的影响。结果益气活血化痰方药中、高剂量组可显著降低COPD大鼠血清IL-8、TNF-α及BALF中MUC5AC的表达水平,其中,高剂量组效果优于中剂量组。结论益气活血化痰方药中、高剂量组能降低COPD大鼠血清中IL-8、TNF-α和BALF中MUC5AC的表达水平,抑制气道炎性反应和缓解气道重塑,揭示益气活血化痰法临床疗效可能的作用机制,为临床治疗提供科学依据。Objective To observe the effects of therapy of benefiting Qi, activating blood, resolving phlegm on cytokines and airway remodeling in chronic obstructive pulmonary disease COPD rats, and to explore its possible therapeutic mechanism for COPD. Methods COPD rat model was established by exposing to cigarette smoking and intratracheal instillation of lipopolysaecharide（LPS）. And then we observed the effects of Chinese herbal medicine with the actions of benefiting Qi, activating blood, resolving phlegm on the expression levels of tumor necrosis factor alpha （TNF-α）, interleukin-8 （IL-8） in the serum of rats, MUC5AC in broncho-alveolar lavage fluid（BALF）, and the pathological changes of bronchus and lung tissues. Results Middle and high dose of Chinese herbal medicine could significantly reduce the expression levels of IL-8, TNF-α in the serum and MUC5AC in BALF of COPD rats, and high dose group showed better effect than middle dose group. Conclusion Middle dose and high dose of Chinese herbal medicine with the actions of benefiting Qi, activating blood, resolving phlegm can decrease the expression levels of IL-8 and TNF-α in the serum and MUC5AC in BALF, inhibit airway inflammation and alleviate airway remodeling, which probably contribute to the therapeutic mechanism.
