摘 要:目的观察小半夏汤对化疗性异食癖大鼠多巴胺(DA)合成与代谢的影响,探讨小半夏汤防治化疗性恶心呕吐的作用机制。方法取Wistar大鼠108只,随机分为6组,即正常对照组,小半夏汤正常对照组(1.6g·kg-1),顺铂模型组,昂丹司琼组(2.6 mg·kg-1),小半夏汤大、小剂量组(3.2,1.6 g·kg-1),每组18只,单笼饲养。适应性饲养7 d后,造模前4 d投放高岭土颗粒。正常对照组和顺铂模型组给予等容量蒸馏水,灌胃给药,每天2次,首次给药后1 h顺铂模型组和各治疗组动物腹腔注射6 mg·kg-1顺铂建立大鼠化疗性异食癖模型。正常对照组和小半夏汤正常对照组腹腔注射等容积生理盐水。造模后每12 h称量并记录动物高岭土摄入量。分别于造模后24 h和72 h取材,测定大鼠血清、回肠和延髓DA及其代谢产物3,4-二羟基苯乙酸(DOPAC)含量以及回肠和延髓酪胺酸羟化酶(TH)、单胺氧化酶B(MAOB)水平。结果顺铂模型组大鼠摄食高岭土量显著升高,小半夏汤大、小剂量组均可抑制化疗大鼠摄食高岭土,均可降低化疗大鼠血清、回肠和延髓DA含量以及回肠、延髓TH水平,对DOPAC含量和MAOB水平没有明显影响。结论小半夏汤防治化疗性恶心呕吐作用机制与降低外周与中枢TH水平、抑制DA合成有关。Objective To observe the effect of Xiao Banxia Tang(XBXT, Minor Rhizoma Pienelliae Decoction) on the synthesis and metabolism of dopamine (DA) in chemotherapy-induced pica rats, and to explore its preventive and therapeutic mechanism for chemotherapy-induced nausea and vomiting. Methods A total of 108 Wistar rats were randomly divided into 6 groups, normal control group, XBXT normal control group (1.6 g .kg-), cisplatin model group, ondansetron group(2.6 mg.kg-1), high- and low-dose XBXT groups(3.2, 1.6 g.kg-1) , 18 rats in each group. The rats were fed solitarily and were given adaptive feeding for 7 days. Four days before the modeling, the rats were given kaolin granules. The normal control group and model group were given distilled water orally, and the rats in other groups were given the corresponding medicine orally twice per day. One hour after the first mediation, rats in the model group and the medication groups were injected intraperitoneally with eisplatin 6 mg" kg-1 to induce chemotherapy-induced pica model, and the rats in the normal control and XBXT normal control group were injected with the equal volume of saline. Every 12 h after the modeling, the intake volume of kaolin was recorded. The contents of DA and dihydroxy-phenyl acetic acid (DOPAC) in rat serum, ileum and medulla oblongata, and the levels of tyramine hydroxylase(TH) and monoamine oxidase-B(MAOB) in the ileum and medulla oblongata were measured 24 h, 72 h after the modeling. Results Kaolin intake in the model group was significantly increased. The high and low dosages of XBXT could significantly inhibit the intake of kaolin in cisplatin-treated rats, reduce DA contents in the serum, ileum and medulla oblongata, and decrease TH and MAOB levels in the ileum and medulla oblongata. However, XBXT showed no effect on DOPAC content and MAOB level.
