摘  要:目的建立高效液相色谱(HPLC)法同时测定复方红根草片中绿原酸、穿心莲内酯和脱水穿心莲内酯含量的方法。方法采用Waters Sun Fire TM C18(4.6 mm×150mm,5μm)色谱柱;流动相为甲醇-0.2%磷酸水溶液,梯度洗脱;流速为1.0 m L·min-1;检测波长:327 nm(0~9 min,绿原酸),225 nm(9~20 min,穿心莲内酯),254 nm(20~45 min脱水穿心莲内酯)。结果绿原酸在0.134~2.425μg范围内线性关系良好(r=0.9999,n=6),平均回收率为99.64%,RSD为1.3%(n=9);穿心莲内酯在0.031~0.553μg范围内线性关系良好(r=0.9999,n=6),平均回收率为99.95%,RSD为1.5%(n=9);脱水穿心莲内酯在0.049~0.887μg范围内线性关系良好(r=0.9998,n=6),平均回收率为99.31%,RSD为1.1%(n=9)。结论该方法操作简单、可靠、准确、重复性好,可用于复方红根草片的质量控制。Objective To simultaneously determine chlorogenic acid, andrographolide and dehydroandrographolide in Compound Honggencao Tablets by HPLC. Methods Waters SunFireTM C18(4.6 mm×150mm,5μm) column was used. The mobile phase consisted of methanol water solution containing 0.2 % phosphoric acid with a gradient elution mode. The flow rate was 1.0 mL. min-1 The detection wavelength was 327 nm(0-9 min) for chlorogenic acid, 225 nm (9-20 min) for andrographolide and 254 nm (20-45min) for dehydroandrographolide. Results Chlorogenic acid had a good linearity in the range of 0.134-2.425 μg, and the average recovery was 99.64 % with RSD of 1.3 %. Andrographolide had a good linearity in the range of 0.031-0.553 μg, and the average recovery was 99.95 % with RSD of 1.5 %. Dehydroandrographolidehad a good linearity in the range of 0.049-0.887 μ g, and the average recovery was 99.31% with RSD of 1.1%. Conclusion The method is simple, accurate and reproducible. It can be used for the quality control of Compound Honggencao Tablets.
