摘 要:目的优选通络调脂胶囊的水提工艺。方法采用正交试验设计,用高效液相色谱法(HPLC)同时测定黄芩苷、芍药苷、葛根素3种指标成分的含量。色谱柱:Agilent ZORBAX Eclispse SB-C18柱(4.6 mm×250 mm,5μm);色谱条件:流动相乙腈-0.1%磷酸水梯度洗脱,流速1.0 m L·min-1,检测波长为280 nm(黄芩苷)、230nm(芍药苷)、250 nm(葛根素)。结果根据回归方程黄芩苷、芍药苷和葛根素分别在0.3336~2.0016μg·m L-1、0.1322~0.7932μg·m L-1、0.0938~0.5628μg·m L-1范围内进样量与峰面积之间线性关系均良好;3个成分的平均回收率分别为100.9%、103.0%和98.3%。最佳提取工艺为加6倍量水,提取3次,每次1 h;工艺验证黄芩苷、芍药苷、葛根素转移率分别为92.83%、87.98%、86.83%,浸膏得率为33.80%。结论采用多指标综合评分法优选通络调脂胶囊的提取工艺,方法简便,准确可靠。Objective To optimize the extraction process of Tongluo Tiaozhi Capsules. Methods Orthogonal design was adopted for the muhiple-index evaluation and extraction process optimization. HPLC was developed for determining baicalin, paeoniflorin, and puerarin in Tongluo Tiaozhi Capsules. Chromatographic column was Agilent ZORBAX Eclispse SB-C18(4.6 mm×250 mm,5μm). The mobile phase consisted of acetonitrile-0.1% phosphoric acid with gradient elution at the flow rate of 1.0 mL .min-1. The detection wavelength was 280 nm for baicalin, 230 nm for paeoniflorin and 250 nm for puerarin. Results The optimal extraction technology was as follows: extraction with 6-fold water for three times, one hour for each extraction. The amount of baicalin, paeoniflorin, puerarin was 92.83 %, 87.98 %, 86.83 %, respectively, and the extraction rate was 33.80 %. The baicalin, paeoniflorin and puerarin all had good linear relationship in the ranges of 0.3336-2.0016 μg, 0.1322-0.7932 μg, 0.0938-0.5628μg, and the average recovery was 100.9 %, 103.0 %, 98.3 %, respectively. Conclusion The established method is simple, accurate, reproducible, and effective, can be used for the quality evaluation of Tongluo Tiaozhi Capsules.
