摘 要:糖激酶是一类利用ATP中的磷酸基团使糖磷酸化的酶,产生的磷酸糖可进一步转化为糖核苷酸(NDP-糖),为自然界中糖类的合成代谢提供糖基供体.糖激酶不仅在植物、动物和微生物的很多生理及病理过程中起重要作用,也是合成糖类化合物的催化剂及药物开发的靶点,研究其活性既有理论意义又有应用价值.然而,糖激酶催化的反应产物磷酸糖一般不含易检测基团,直接检测并不容易,因此,许多方法被开发用来检测糖激酶的活性.综述了糖激酶活性检测技术的最新进展.Sugar kinases are enzymes that transfer phosphate groups from ATPs to sugars to form sug- ar phosphates, which can be further converted into sugar nucleotides (NDP-sugars), providing sugar donors for carbohydrates synthesis in nature. Sugar kinases not only play critical roles in a variety of physiological and pathological processes in plants, animals and microbes, but are also catalysts of carbohydrate synthesis and targets for drug development. Studies on the activity of sugar kinases are of both theoretical and practi- cal significance, However,since sugar phosphates,the product of the reactions catalyzed by sugar kinases, normally do not contain groups easy to detect, direct monitor of their production becomes difficult. There- fore,many methods have been developed for sugar kinase activity assay. This paper reviews the up-to-date techniques for sugar kinase activity assay.
