摘 要:目的建立刺五加、五加皮、红毛五加皮中原儿茶酸、紫丁香苷、绿原酸的质量分数测定方法。方法采用HPLC法,色谱柱为Kromasil C18色谱柱;流动相为乙腈-0.1%(体积分数)H3PO4溶液,梯度洗脱;检测波长为270 nm;流速为1.0 m L/min;柱温为30℃。结果刺五加中原儿茶酸、紫丁香苷、绿原酸达到基线分离,线性关系良好(r≥0.999 9),平均加样回收率分别为98.7%、99.7%、101.0%,RSD均小于3.0%(n=6)。刺五加、五加皮、红毛五加皮中原儿茶酸质量分数分别为0.022 5~0.028 7、0.076 3~0.132、0.013 8~0.071 2 mg/g;紫丁香苷质量分数分别为0.445~0.616、0.023 8~0.025 9、0.037 3~0.069 3 mg/g;绿原酸质量分数分别为1.834~2.169、0.659~0.713、0.372~0.709 mg/g。刺五加中紫丁香苷、绿原酸质量分数最高,其中紫丁香苷约为五加皮、红毛五加皮中质量分数的7~25倍,绿原酸约为五加皮、红毛五加皮中质量分数的3~5倍;而原儿茶酸在五加皮中质量分数最高,其次为红毛五加皮、刺五加。结论该方法灵敏、可靠,可用于刺五加、五加皮和红毛五加皮类药材质量控制的参考。Objective To establish a determintation method for protocatechuic acid,syringing and chlorogenic acid in Acanthopanax senticosi Radix et Rhizoma seu Caulis,Acanthopanacis Cortex and Acanthopanax giraldii Cortex by HPLC. Methods HPLC method was performed with the Kromasil C18 column. The acetonitrile-0.1% phosphoric acid( gradient elution) was employed as a mobile phase,and the detection wavelength was 270 nm. Column temperature was 30 ℃,and the flow rate was 1. 0 m L / min.Results Baseline separation was obtained and the linear relationship was good( r≥0.999 9) for the three constituents. The average recoveries of protocatechuic acid,syringin,and chlorogenic acid were 98. 7%,99. 7% and 101.0%,and their RSD were below 3.0%( n = 6). In Acanthopanax senticosi Radix et Rhizoma seu Caulis,Acanthopanacis Cortex and Acanthopanax giraldii Cortex,the content of protocatechuic acid was0.022 5- 0.028 7,0.076 3- 0.132,0.013 8- 0.071 2 mg / g,and the content of syringin was 0.445- 0.616,0. 023 8- 0.025 9,0.037 3- 0.069 3 mg / g,and that of chlorogenic acid was 1.834- 2.169,0.659- 0.713,0. 372- 0.709 mg / g,respectively. The contents of syringin and chlorogenic acid in Acanthopanax senticosi Radix et Rhizoma seu Caulis were the highest,in which that of syringin was as about 7- 25 times high as canthopanacis Cortex and Acanthopanax giraldii Cortex while that of chlorogenic was as 3- 5 times. The content of protocatechuic acid in Acanthopanacis Cortex was the highest,followed by Acanthopanax giraldii Cortex and Acanthopanax senticosi Radix et Rhizoma seu Caulis. Conclusion The method is sensitive,reliable and available for the quality control of Acanthopanax senticosi Radix et Rhizoma seu Caulis,Acanthopanacis Cortex and Acanthopanax giraldii Cortex.
