摘  要:目的研究毛蕊花糖苷对高脂模型大鼠血管内皮细胞凋亡相关蛋白的表达作用。方法高脂饮食喂养建立高血脂模型,设模型组、辛伐他汀组(10 mg/kg)、毛蕊花糖苷低剂量组(50 mg/kg)、毛蕊花糖苷中剂量组(100 mg/kg)、毛蕊花糖苷高剂量组(200 mg/kg)和空白对照组,利用生化分析仪测定血脂、抗氧化指标,采用Western-blot法测定腹主动脉中细胞凋亡相关蛋白基因Bax与Bcl-2的表达。结果与模型组比较,毛蕊花糖苷高剂量组能显著降低模型大鼠血清总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)水平与脂质过氧化产物(MDA)含量(P<0.01),总超氧化物歧化酶(T-SOD)含量与高密度脂蛋白胆固醇(HDL-C)水平明显提高(P<0.01或P<0.05)。毛蕊花糖苷中、高剂量组Bax蛋白表达显著下调、Bcl-2蛋白表达明显上升(P<0.01)。结论毛蕊花糖苷通过调节凋亡蛋白Bax-Bcl-2,对抗高脂模型大鼠所致的血管内皮细胞凋亡及高脂血症所致的氧化损伤。Objective To study the effects of verbascoside on the expression of apoptosis-related proteins in vascular endothelial cells in rats with hyperlipidemia. Methods The rat models with hyperlipidemia were established by feeding high-lipid diets and then were randomly divided into the model group,the simvastatin group( 10 mg / kg),three verbascoside groups with low,middle and high dosages( 50,100 and 200 mg /kg),and the control group. The blood lipids and antioxidant index were determined by biochemical analyzer. The expression of apoptosis-related protein Bax and Bcl-2 in abdominal aorta was determined by western blot. Results Compared to the model group,the high dosage verbascoside group significantly reduced the levels of serum total cholesterol( TC),glycerol lipid( TG),low density lipoprotein cholesterol( LDL-C) and lipin peroxide( MDA)( P0.01),and increased the levels of total superoxide dismutase( T-SOD) and high density lipoprotein cholesterol( HDL-C)( P0.05 or P0.01). The expression of Bax in middle and high dosage groups was significantly decreased,but Bcl-2 expression was increased( P〈0. 01). Conclusion Verbascoside can reduce the apoptosis of vascular endothelial cells and oxidative damage in rats with hyperlipidemia through regulation of Bax and Bcl-2 expression.
