摘 要:目的探讨简单有效的左心疾病相关肺动脉高压(PH-LHD)大鼠模型的制备方法。方法雄性成年Wistar大鼠进行主动脉弓缩窄(transverse aortic constriction,TAC)手术,9周后检测大鼠左心室舒张末压、股动脉压、心输出量,并通过组织HE染色观察肺动脉血管的重构,通过免疫荧光染色观察肺间质Ⅰ型胶原的变化。结果主动脉弓缩窄9周后,大鼠的左心室舒张末压显著增加,心输出量、心率均显著降低,肺血管发生显著重构,管腔面积减小,同时肺间质Ⅰ型胶原显著增加。结论通过主动脉弓缩窄术成功制作了大鼠PH-LHD模型。Objective To explore a simple and effective model of pulmonary arterial hypertension due to left heart disease( PH-LHD) in rats. Methods Adult male Wistar rats were performed by transverse aortic constriction surgery. After 9 weeks,left ventricular end diastolic pressure,femoral artery pressure,cardiac output and heart rate were chronologically collected. The pulmonary artery vascular remodeling was observed by hematoxylin-eosin staining. The pulmonary interstitial collagen Ⅰ was observed by immunofluorescent staining. Results Compared with the sham group,the left ventricular end diastolic pressure was obviously increased,but the cardiac output and heart rate were significantly decreased in the model group. The vascular remodeling and reduced pulmonary artery lumen size were observed in the model group. Pulmonary interstitial collagenⅠwas increased in the model group. Conclusion A stable PH-LHD rat model could be successfully established by transverse aortic constriction in this research.
